摘  要:本试验以黄秋葵叶片和带节茎段为外植体,采用组织培养的方法,研究不同激素组合对愈伤组织诱导、愈伤组织增殖、不定芽分化、生根的影响,筛选出适合各阶段的最佳培养基配方,同时对不同种类种子消毒剂的消毒效果进行探究。结果表明,黄秋葵种子最佳消毒方法为75%C2H5OH浸泡2 min+0.15%HgCl2浸泡20 min;愈伤组织诱导最佳培养基为MS+2.0 mg/L 6-BA,带节茎段的诱导率达98%;增殖最佳培养基为MS+2.0 mg/L 6-BA+0.2 mg/L NAA,增殖系数为4;分化最佳培养基为MS+2.0 mg/L ZT+0.15 mg/L NAA,分化率为93%;生根最佳培养基为MS+0.1 mg/L NAA,主根粗壮,根系发达,质量好。本试验完善了黄秋葵组织培养及再生体系,为其种质资源的保护、利用、基因改造和其他植物的组织培养研究提供了科学依据。In this experiment,the leaves and stem segments of Abelmoschus esculentus L.were used as explants,and the effects of different hormone combinations on callus induction,callus proliferation,adventitious bud differentiation and rooting were studied by tissue culture.The best medium formula suitable for each stage was selected,and the disinfection effect of different disinfectants was briefly explored.The results showed that the best disinfection method was 75%C 2H 5OH soaking for 2 minutes+0.15%HgCl 2 soaking for 20 minutes.The best medium for callus induction was MS+2.0 mg/L 6-BA,and the induction rate of stem segments with nodes reached 98%.The optimal medium for proliferation was MS+2.0 mg/L 6-BA+0.2 mg/L NAA,and the proliferation coefficient was 4.The best differentiation medium was MS+2.0 mg/L ZT+0.15 mg/L NAA,and the differentiation rate was 93%.The best rooting medium:MS+0.1 mg/L NAA,the root quality was good,the main root was strong,and the root system was developed.This experiment improved the process of tissue culture and regeneration system establishment of Abelmoschus esculentus L.,and contributed reference value for the protection,utilization,genetic transformation of its germplasm resources and tissue culture research of other plants.
