摘 要:目的观察辣椒辣素(CAP)对BV2小胶质细胞迁移能力的影响,探讨辣椒辣素镇痛作用的可能机制。方法2021年3月至2022年6月期间,实验动物来源于广州辉园动物实验中心。用脂多糖(LPS)活化小胶质细胞,构建BV2小胶质细胞模型,造模后加入不同浓度的CAP,设置对照组、LPS组及LPS+CAP组,CAP浓度分别为10μmol和25μmol。对照组BV2细胞不做任何处理连续培养48 h;LPS组BV2细胞加入1μg/ml LPS作用24 h后换液,继续培养24 h;LPS+CAP组BV2细胞先加入LPS作用24 h后换液,再加入不同浓度CAP处理24 h,分别为LPS+10μmol CAP组及LPS+25μmol CAP组。经消化、离心、洗涤等步骤后取下室细胞显微镜下染色、拍照、计数,观察不同浓度的CAP对LPS激活的小胶质细胞迁移能力的影响。结果两种浓度的CAP均减少LPS活化的BV2细胞迁移数量,高浓度的CAP对BV2细胞迁移数量影响更大,不同浓度CAP对LPS诱导的小胶质细胞的活化有不同程度的抑制作用,与LPS组(113.7±2.1)比较,低浓度组和高浓度组CAP可以减少LPS活化的BV2细胞迁移数量分别为(90.0±11.4和77.7±8.6),差异具有统计学意义(P均<0.05)。结论CAP可以通过影响小胶质细胞的迁移能力,抑制小胶质细胞的激活,减少由此引起的炎症介质释放而达到镇痛作用。Objective To investigate the effect of capsaicin(CAP)on migration of BV2 microglial cell and explore the possible analgesic mechanism of capsaicin.Methods From March 2021 to June 2022,the experimental animals were sourced from Guangzhou Huiyuan Animal Experimental Center.Microglia cells were activated by lipopolysaccharide(LPS),and the model of BV2 microglia cells was constructed.After the model was established,capsaicin with different concentrations was added into the model,control group,LPS group and LPS+CAP group,with 10μmol and 25μmol.BV2 cells were cultured in the control group for 48 h without any treatment,and were treated with 1μg/ml LPS in LPS group for 24 h,then cultured for 24 h.The BV2 cells were treated with LPS in LPS+CAP group for 24 h,then that with different concentrations of CAP for 24 h in LPS+10μmol CAP group and LPS+25μmol CAP group.The effect of capsaicin with different concentrations on the migration ability of LPS-activated microglial cells was observed under the microscope.Results Both 10μmol and 25μmol concentration of CAP reduced the migration number of LPS-activated BV2 cells,and the high concentration had a greater effect on the number of BV2 cell migration.Different concentrations of CAP inhibited the activation of LPS-induced microglia cells to different degrees,in the high concentration LPS group(113.7±2.1),in low concentration group(90.0±11.4,77.7±8.6)respectively,with significant statistic difference(all P<0.05).Conclusion Capsaicin can inhibit the activation of microglia and reduce the release of inflammatory mediators by affecting the migration ability of microglia.
