摘 要:马头山羊是武陵山脉的地方良种,以个体较大,肉和板皮质量优良而著称,是湖北省重点保护的品种。本研究选取性别不同的1岁马头山羊作为试验对象,利用纤维蛋白酶消化法建立马头山羊耳部组织成纤维细胞系,并分析、比较成纤维细胞不同天数生长形态、生长曲线和冻存前后存活率等生物学特征。结果表明,培养48 h后,可观察到组织块周围有许多典型成纤维细胞形态的细胞环绕生长,细胞倍增时间约为24 h,细胞冻存后的复苏率分别为96.0%和96.5%,细胞生长呈现出典型的“S”形。此细胞系的建立为马头山羊遗传资源保护及其他研究提供理想的试验材料。Matou goat is a local excellent breed in the Wuling Mountains,known for its large size,excellent meat and skin quality,and is a key protected variety in Hubei Province.This study selected 1-year-old Matou goats of different genders as the experimental subjects,established afibroblast cell line of Matou goat ear tissue usingfiber protease digestion method,analyzed and compared the biological characteristics such as cell morphology,growth curve,survival rate before and after cryopreservation.The results showed that after 48 hours of cultivation,many typicalfibroblast shaped cells could be observed surrounding the tissue block for growth.The cell doubling time was about 24 hours,and the recovery rates after cell cryopreservation were 96.0%and 96.5%respectively.The cell growth showed a typical“S”shape.The establishment of this cell line provides an ideal experimental material for the protection of genetic resources,and other basic research of Matou goats.
