摘  要:目的研究天麻素在MCAO大鼠脑梗死急性期对炎症的抑制作用及其作用机制。方法将SD大鼠分为正常组、假手术组、模型组及天麻素组,采用MCAO方法将后两组建立大鼠脑梗死模型,天麻素组每天给予腹腔注射天麻素,剂量为100 mg/kg。其余各组腹腔注射相同体积的生理盐水。并在1,3,7d 3个时间点取材,采用Western blot法分析梗死半球内炎性因子NF-κB、TNF-α、INF-γ以及Notch通路相关蛋白Notch1、Hes1、Hes5的含量。结果大鼠进行MCAO造模后1周内,患侧大脑半球内的炎性因子NF-κB、TNF-α、INF-γ均有不同程度的升高(P<0.05),而天麻素能够使其含量降低(P<0.05)。而且,MCAO造模后,大鼠患侧大脑半球中Notch1、Hes1、Hes5蛋白表达明显增高(P<0.05),而天麻素能够使Notch通路相关蛋白进一步升高(P<0.05)。结论天麻素能够抑制MCAO大鼠脑梗死急性期炎性反应,其作用机制可能与调控Notch通路有关。Objective To study the inhibitory effect of gastrodin on inflammation and its mechanism in acute cerebral infarction of middle cerebral artery occlusio(MCAO)rats.Methods SD rats were divided into control group,sham group,model group and gastrodin group.MCAO method was used to establish the rat cerebral infarction model in the latter two groups.Gastrodin group received intraperitoneal injection of gastrodin(100 mg/kg)every day,while the other groups received normal saline in same volume.Samples were collected in day 1,day 3 and day 7.Then Western blot was used to measure the amount of inflammatory cytokines[nuclear transcription factor(NF-κB),tumor necrosis factor-α(TNF-α),interferon-gamma(INF-γ)]and Notch pathway-related proteins[transmembrane receptor protein Notch-1(Notch1),human transcription factor HES1(Hes1),human transcription factor HES1(Hes5)].Results One week after MCAO modeling,inflammatory factors(NF-κB,TNF-α,INF-γ)in ischemic hemisphere were increased in different degrees(P<0.05),while those in gastrodin group decreased(P<0.05).In addition,the level of Notch pathway-related proteins(Notch1,Hes1,Hes5)in the model group increased compared to those in the sham group(P<0.05).Those in the gastrodin group became even higher(P<0.05).Conclusion Gastrodin can inhibit the inflammatory response in acute stage of cerebral infarction in MCAO rats and the mechanism may be related to Notch pathway.
