摘  要:目的探讨十溴二苯乙烷(decabromodiphenyl ethane,DBDPE)对大鼠雄性生殖系统的损伤作用及豌豆肽的保护作用。方法SPF级成年雄性Wistar大鼠40只,随机分为对照组、DBDPE低剂量组、DBDPE高剂量组和DBDPE高剂量+豌豆肽组。DBDPE和豌豆肽分别经口灌胃和饮水给予大鼠,连续8周。实验结束,采用精子分析仪分析大鼠精子活力,精子快速染色和苏木素-伊红(HE)染色分别评价大鼠精子形态和睾丸结构改变,免疫组织化学染色和Western blot检测生殖细胞特异标记蛋白死盒解旋酶4(DEAD-box polypeptide 4,Ddx4)蛋白表达,TUNEL染色检测睾丸细胞的凋亡,Western blot检测睾丸组织凋亡相关蛋白Bax、Bcl2、Caspase-3和cleaved Caspase-3的蛋白表达。结果与对照组相比,DBDPE染毒组大鼠精子活力与质量明显下降(P<0.05),染毒组大鼠精子出现畸形;染毒大鼠睾丸生精小管生精上皮厚度降低、空泡化;染毒大鼠生殖细胞Ddx4表达降低;染毒大鼠睾丸生殖细胞凋亡;DBDPE染毒大鼠睾丸组织Ddx4蛋白表达水平较对照组明显降低(LSD-t=4.85,LSD-t=6.90,LSD-t=2.94,P<0.05),Bax和cleaved Caspase-3蛋白表达水平升高,Bcl2表达下降;与DBDPE高剂量组相比,DBDPE高剂量+豌豆肽组的精子活力降低、睾丸形态学损伤在一定上得到改善,大鼠生殖细胞凋亡明显减轻。结论DBDPE导致雄性大鼠睾丸组织病理损伤、生殖细胞凋亡、精子活力下降,豌豆肽对DBDPE的生殖毒性具有一定的保护作用。ObjectiveeTo investigate the male reproductive toxicity induced by decabromodiphenyl ethane(DBDPE)and the protective effects of pea peptide in rats.Methods Forty SPF adult male Wistar rats were randomly divided into control group,low-dose DBDPE group,high-dose DBDPE group,and high-dose DBDPE+pea peptide group.DBDPE and pea peptide were administered to the rats by gavage and drinking water,respectively,for 8 consecutive weeks.Ater cessation of the experiment,sperm vitality was analyzed using a sperm analyzer.The rapid sperm staining and hematoxylin-eosin(H-E)staining were employed to evaluate the morphologic changes of sperm and testicular structure.The protein expression of DEAD-box polypeptide 4(Ddx4),germ cell-specific marker,was detected by both of immunohistochemical staining and Western blot.The apoptosis of germ cells was revealed by TUNEL staining.In addition,proteins expression of apoptosis-related proteins including Bax,Bcl2,Caspase-3 and cleaved Caspase-3 were examined by Western blot.Results Compared with the control group,the sperm vitality and quality of the DBDPE-treated rats were significantly reduced(P<0.05),with the sperm abnormality.The thickness of the spermatogenic epithelium of the seminiferous tubules in DBDPE-treated rats became thinner than that of control rats,and vacuolar degeneration in epithelium cells were observed.The expression of Ddx4 in germ cells of DBDPEtreated rats was decreased(LSD-t=4.85,LSD-t=6.90,LSD-t=2.94,P<0.05),and the significant apoptosis of germ cells in the testes were also revealed by TUNEL staining in DBDPE-treated rats.The levels of Bax and cleaved Caspase-3 proteins increased,while Bcl2 expression decreased.The decrease of sperm vitality and morphological damage of testes were reversed to some extent by pea peptide.The pea peptide also significantly alleviated the apoptosis of germ cells induced by DBDPE.Conclusion DBDPE exposure to male rats result ed in obvious reproductive toxicity characterized by pathological damage of testicular tissue,apoptosis of germ ce
