摘 要:为明确香榧根腐病病原菌的种类,本研究采用常规组织分离法分离纯化病原菌,根据柯赫氏法则对病原菌进行致病性检测,通过形态学和分子生物学鉴定确定病原菌种类。对香榧病株进行常规组织分离,分离纯化得到12个菌株,经分子生物学鉴定将这12个菌株归为10类,分别用ZJ-1至ZJ-10表示;将ZJ-1至ZJ-10进行致病性检测,根据柯赫氏法则,证实ZJ-10是香榧根腐病的病原菌;结合菌株形态学特征和ITS rDNA序列,可将ZJ-10菌株鉴定为腐皮镰孢菌(Fusarium solani)。即香榧根腐病的病原菌是腐皮镰孢菌。Clarify the type of pathogen of T.grandis 'Merrillii' root rot disease.The isolated pathogens were tested for pathogenicity and the classification status of the pathogens was determined by using molecular techniques combined with morphological identification.T.grandis 'Merrillii' disease strains were isolated from tissues,and 12strains were isolated and purified.After molecular biological identification,these 12 strains were classified into 10categories,represented by ZJ-1 to ZJ-10.The pathogenicity test of ZJ-1 to ZJ-10 was carried out,and according to Kocher's law,it was confirmed that ZJ-10 is the pathogen of T.grandis 'Merrillii' root rot disease.ZJ-10 strain was identified as Fusarium solani by combining morphological characteristics and ITS rDNA sequence.The pathogen causing T.grandis 'Merrillii' root rot is Fusarium solani.
