水药麻骨风醇提液对类风湿关节炎大鼠模型血清IL-6、TNF-ɑ水平以及踝关节软组织VEGF、MMP-3表达的影响

摘　　要：目的:观察麻骨风醇提物对牛II型胶原诱导关节炎大鼠血清TNF-ɑ、IL-6和踝关节软组织VEGF、MMP-3阳性表达的影响,初步探讨麻骨风治疗类风湿关节炎的药理机制。方法:采用牛Ⅱ型胶原诱导大鼠关节炎模型,将60只SD大鼠随机分为空白组、模型组和麻骨风醇提取物低、中、高剂量组,每组10只。初次致炎24 h后,除空白组和模型组外,给予灌胃给药1次/d,连续给药4周,采用ELISA法测定IL-6、TNF-ɑ水平,免疫组化法测定踝关节软组织VEGF、MMP-3的阳性表达。结果:与空白组比较,模型组大鼠血清IL-6、TNF-ɑ的含量和踝关节软组织VEGF、MMP-3的阳性表达明显增高(P<0.05);与模型组比较,低剂量组、中剂量组、高剂量组大鼠IL-6、TNF-ɑ的含量和踝关节软组织VEGF的阳性表达明显降低(P<0.05),中剂量组、高剂量组大鼠踝关节软组织MMP-3的阳性表达明显降低(P<0.05)。结论:水药麻骨风可以降低RA大鼠血清IL-6、TNF-ɑ的含量和踝关节软组织VEGF、MMP-3的阳性表达,起到抑制炎症因子释放,缓解RA的发展进程。Objective:To observe the effects of the alcohol extract of Gnetem parvifolium on serum levels of TNF-ɑand IL-6,as well as the expression of VEGF and MMP-3 in the soft tissue of the ankle joint in rats with type II collagen-induced arthritis.This study aims to preliminarily explore the pharmacological mechanism of Gnetem parvifolium in treating rheumatoid arthritis.Methods:Using a rat model induced by type II collagen,60 SD rats were randomly divided into a blank group,a model group,and three groups of low,medium,and high doses of the alcohol extract of Gnetem parvifolium,with 10 rats in each group.After initial inflammation for 24 hours,except for the blank and model groups,rats were administered orally once daily for four weeks.The levels of IL-6 and TNF-ɑwere measured using ELISA,and the expression of VEGF and MMP-3 in the soft tissue of the ankle joint was determined using immunohistochemical methods.Results:Compared to the blank group,the levels of IL-6 and TNF-ɑin serum and the expression of VEGF and MMP-3 in the soft tissue of the ankle joint in the model group significantly increased(P<0.05).Compared to the model group,the levels of IL-6 and TNF-ɑin serum and the expression of VEGF in the soft tissue of the ankle joint in rats from the low,medium,and high dose groups significantly decreased(P<0.05),and the expression of MMP-3 in the soft tissue of the ankle joint in rats from the medium and high dose groups significantly decreased(P<0.05).Conclusion:The alcohol extract of Gnetem parvifolium can reduce serum levels of IL-6 and TNF-ɑas well as the expression of VEGF and MMP-3 in the soft tissue of the ankle joint in rats with rheumatoid arthritis.This suggests that it can inhibit the release of inflammatory factors and alleviate the progression of RA.
