摘  要:目的 探讨松果菊苷对便秘小鼠的通便作用及其机制。方法 KM小鼠分为对照组、模型组和松果菊苷低、高剂量组(30、70 mg/kg),每组8只,除对照组外,其余各组小鼠用洛哌丁胺灌胃法构建便秘模型。记录小鼠排便次数、粪便粒数和粪便含水量,钡餐推进法评估小肠转运率,HE染色观察结肠组织形态,试剂盒检测结肠组织SOD、GSH-Px活性及IL-1β、TNF-α水平,免疫荧光法检测结肠组织AQP3蛋白表达,Western blot法检测结肠组织SR4、SP、VIP、AQP3、CFTR、PKA蛋白表达。结果 与模型组比较,松果菊苷各剂量组小鼠排便频率、粪便颗粒数、粪便含水量、肠道转运率,以及结肠组织SOD、GSH-Px活性和SR4、SP、AQP3、CFTR、PKA蛋白表达升高(P<0.05,P<0.01),结肠组织病理损伤得到改善,IL-1β、TNF-α水平和VIP蛋白表达降低(P<0.05,P<0.01)。结论 松果菊苷对便秘小鼠具有通便作用,可能与减轻肠道氧化应激及炎症反应、改善肠神经递质异常和增加结肠组织AQP3表达有关。AIM To investigate the laxative effects of echinacoside on constipation mice and the mechanisms.METHODS KM mice divided into the control group,the model group and the low and high doses echinacoside groups(30,70 mg/kg),with 8 mice in each group,were induced into constipation models by gavage feeding of loperamide except for those of the control group.The mice had their defecation frequency,counts of fecal particles and fecal water content recorded;their rate of small intestine transport evaluated by barium meal propulsion method;their colonic morphology observed by HE staining;their colonic activities of SOD,GSH-Px and the levels of IL-1βand TNF-αdetected by kit;their colonic AQP3 protein expression detected by immunofluorescence;and their colonic protein expressions of SR4,SP,VIP,AQP3,CFTR and PKA detected by Western blot.RESULTS Compared with the model group,both echinacoside groups displayed increased defecation frequency,counts of fecal particles,fecal water content,intestinal transport rate,colonic activities of SOD and GSH-Px,protein expressions of SR4,SP,AQP3,CFTR and PKA(P<0.05,P<0.01);improved colonic pathological injury;and decreased levels of IL-1β,TNF-α,and VIP protein expression(P<0.05,P<0.01).CONCLUSION Being a laxative to constipation mice,echinacoside may prompt reduction of intestinal oxidative stress and inflammatory reaction via improvement of intestinal neurotransmitter abnormality and colonic AQP3 expression.
