摘 要:以半红树植物小草海桐叶片为外植体,MS为基础培养基,设计不同质量浓度浓度的6-BA、NAA和6-BA等激素,探究不同阶段小草海桐组培技术的最适宜浓度。结果表明,愈伤组织诱导阶段MS+0.5 mg/L NAA+0.1 mg/L 6-BA的培养基效果最好,诱导率为82.01%;在不定芽的诱导阶段,MS+1.0 mg/L 6-BA的培养基效果最好,出芽率为60.42%;在生根培养过程中,MS+0.5 mg/L IAA的培养基效果最好,生根率为87.67%。Using the leaves of the semi-mangrove plant Scaevola hainanensis as explants and MS as the base medium,different concentrations of hormones such as 6-BA,NAA and 6-BA were designed to explore the optimal concentration of tissue culture technology of Scaevola hainanensis in different stages.The results showed that MS+0.5 mg/L NAA+0.1 mg/L6-BA medium had the best effect on callus induction stage,and the induction rate was 82.01%.In the stage of bud induction,MS+1.0 mg/L6-BA medium had the best effect,and the budding rate was 60.42%.In the process of rooting culture,MS+0.5 mg/L IAA medium had the best effect,and the rooting rate was 87.67%.
