“和郁脂”抗抑郁中药脂质体胶囊的研制

摘　　要：“和郁脂”抗抑郁中药脂质体胶囊以贯叶连翘为主药,配以“逍遥散”制成。为优选其制备工艺,采用正交试验法对药材提取工艺及脂质体成型工艺进行了考察。得最优提取工艺为,以无水乙醇为溶剂,采用超声波辅助提取,料液比1∶8,提取时间40 min;脂质体成型工艺为,采用薄膜分散法制备脂质体,膜脂比1∶6、水化温度55℃、药脂比1∶20。经脂质体的粒径、PDI等稳定性评价,表明所得中药脂质体粒径均一,较少发生聚集,稳定性好;经体外释放试验表明,脂质体在0~10 h内释药量逐渐递增,12 h后释药速度趋于稳定。“Heyu Zhi”antidepressant liposomal capsule is made of forsythia perforandum as the main drug,with“Xiaoyaoshan”.In order to optimize its preparation process,the orthogonal test method was used to investigate the extraction process of medicinal materials and the liposome molding process.The optimal extraction process was to use absolute ethanol as solvent and ultrasonic assisted extraction,with a material-liquid ratio of 1∶8 and an extraction time of 40 min.The liposome molding process was prepared by film dispersion method,with a membrane-lipid ratio of 1∶6,a hydration temperature of 55℃,and a drug-lipid ratio of 1∶20.The stability evaluation of liposome particle size and PDI showed that the obtained traditional Chinese medicine liposomes had uniform particle size,less aggregation and good stability.In vitro release assays showed that the amount of liposome release gradually increased within 0~10 h,and the release rate tended to be stable after 12 h.
