摘 要:以四倍体资阳香橙幼芽和茎段为外植体,通过微扦插技术,对不同植物激素组合进行了优化,以期建立高效的快速繁殖体系。结果表明:以幼芽为外植体,添加1.0 mg/L 6-BA+0.1 mg/L KT的MT培养基,诱导芽效果最佳,45 d时诱导率高达91.67%,增殖系数为1.82;以茎段为外植体,添加1.0 mg/L 6-BA+1.0 mg/L KT的MT培养基,腋芽分化效果最佳,45 d时腋芽分化率为76.67%,增殖系数为1.22;而2.0 mg/L NAA+0.2%AC(活性炭)的1/2MT培养基,诱导生根的效果最佳,60 d时生根率达92.59%;对获得的46株组培苗进行移栽驯化,存活率为95.7%。经根尖染色体压片计数法鉴定,获得的组培苗均为四倍体资阳香橙。该研究建立了高效的四倍体资阳香橙无性繁殖体系,可为后续多倍体研究和生产实践提供材料。Using tetraploid Citrus.junos cv.Ziyang orange bud and stem segments were used as explants,different plant hormone combinations were optimized through micro cutting technology to establish an efficient rapid propagation system.The results shows that the germination proliferation is the best when MT medium with 1.0 mg/mL 6-BA+0.1 mg/mL KT and the proliferation rate is up to 91.67%and the proliferation coefficient is 1.82 after 45 days.With stem segments as explants,MT medium with 1.0 mg/mL 6-BA+1.0 mg/mL KT hads the best axillary bud differentiation,the axillary bud differentiation rate is 76.67%and proliferation coefficient is 1.22 after 45 days.The 1/2 MT medium with 2.0 mg/mL NAA+0.2%AC(activated carbon)has the best effect on inducing rooting,with a rooting rate of 92.59%after 60 days.46 seedlings are transplanted and domesticated with the result of 95.7%survival rate.With the root tip chromosome compression counting method,the tissue cultured seedlings obtained are tetraploid Citrus.junos cv.Ziyang orange.A high-efficiency sexual reproductive system for tetraploid Citrus.junos cv.Ziyang orange is built in this research,providing a reference for subsequent polyploid research and production practice.
