摘 要:目的观察创面消毒凝胶对大鼠皮肤创伤后愈合的促进作用,并利用网络药理学探讨其潜在作用机制。方法将48只大鼠采用随机数字表法分为空白组、模型组和给药组,每组16只。除空白组外,其余2组建立皮肤创伤模型,造模后给药组在伤口涂抹创面消毒凝胶(1.0 mg/mm^(2)),空白组和模型组涂抹等体积的赋形剂,共涂抹28 d。于给药后第4,7,14,28天计算各组大鼠创面残留率;取造模处皮肤组织,行HE染色,观察各组大鼠皮肤病理变化。通过Cytoscape 3.7.2构建药物-成分-靶点-疾病网络,寻找核心成分及作用靶点,并利用AutoDock进行分子对接验证。结果给药后第4天给药组大鼠伤口开始结痂,给药后第7天模型组大鼠伤口开始结痂;与空白组比较,给药后第4,7,14,28天模型组大鼠创面残留率均明显升高,差异有统计学意义(P<0.01);与模型组比较,给药后第7,14,28天给药组大鼠创面残留率均明显降低,差异有统计学意义(P<0.05或P<0.01)。空白组大鼠皮肤组织真皮层和表皮层完整,少有炎症细胞。模型组大鼠表皮层增厚,真皮层内有大量炎症细胞,给药后第7,14,28天给药组毛细血管数量增多,开始形成完整的表皮基底层;给药后第4,7,14,28天给药组炎症细胞均明显减少(P<0.05或P<0.01)。网络药理学研究结果显示,创面消毒凝胶促进皮肤愈合的关键成分为槲皮素、芹菜素、熊果酸,关键作用靶点为环加氧酶2(PTGS2)、类固醇受体激活蛋白2(NCOA2)、环加氧酶1(PTGS1)。分子对接结果提示,熊果酸与PTGS1、PTGS2、NCOA2的结合能均较低,结合稳定。结论创伤消毒凝胶对破损皮肤愈合具有明显促进作用,其潜在机制与熊果酸等成分调控PTGS1、PTGS2、NCOA2等炎症因子有关。Objective To observe the promotion effect of wound disinfection gel on skin wound healing in rats,and explore its potential mechanism by using network pharmacology.Methods 48 rats were divided into blank group,model group and drug administration group by random number table,with 16 rats in each group.Skin wound model was established in the 2 groups except the blank group.After modeling,wound disinfection gel(1.0 mg/mm^(2))was applied to the wound in the drug administration group,and excipients of equal volume were applied to the blank group and the model group for a total of 28 days.The residual wound rate was calculated on the 4th,7th,14th and 28th day after administration.HE staining was performed to observe the pathological changes of the skin in each group.The drug-composition-target-disease network was constructed by Cytoscape 3.7.2,the core components and targets were searched,and the molecular docking was verified by AutoDock.Results On the 4th day after administration,the wound of rats in the administration group began to form scab,and on the 7th day after administration,the wound of rats in the model group began to form scab.Compared with the blank group,the wound residual rate in the model group was significantly increased at the 4th,7th,14th and 28th days after administration,with statistical significance(P<0.01).Compared with the model group,the wound residual rate of rats in the admi-nistration group was significantly decreased at the 7th,14th and 28th days after administration,with statistical signi-ficance(P<0.05 or P<0.01).The dermis and epidermis of skin tissue of rats in blank group were intact,with few inflammatory cells.The epidermal layer of rats in the model group was thickened,and there were a large number of inflammatory cells in the dermis.On the 7th,14th and 28th days after administration,the number of capillaries in the administration group increased,and the complete basal layer of the epidermis began to form.On the 4th,7th,14th and 28th days after administration,the inflammatory cells
