摘  要:[目的]探析五大连池火山区露头泉细菌和古菌的群落结构及其对扰动的响应。[方法]采集五大连池露头泉水样,按扰动程度将其分为农田组(A1,A2,A3)、村屯组(B1,B2,B3)、景区组(C1,C2,C3)和野外组(D1,D2,D3),利用高通量测序技术对各组别露头泉中细菌和古菌的群落结构进行测序分析。[结果]各露头泉之间菌群多样性存在显著差异,野外组露头泉的细菌和古菌多样性及其丰富度较高;露头泉微生物各物种互作关系网络复杂,但多以Proteobacteria、Actinobacteria和Bacteroidetes等为优势细菌门,以Parvarchaeota、Woesearchaeota和Verrucomicrobia等为优势古菌门;在不同程度的扰动影响下,某些露头泉出现了特异优势菌,如Helicobacteraceae、Verrucomicrobiaceae和Deferribacteres等。[结论]五大连池火山区露头泉分布着丰富的细菌和古菌,人为活动产生的不同程度扰动,导致露头泉中细菌和古菌的多样性、组成及丰度呈现差异化。[Objective]To investigate the community structure of bacteria and archaea and their response to disturbance in spring of Wudalianchi volcanic area,NE China.[Method]Water samples obtained from 12 springs were divided into 4 groups,including farmland(A1,A2,A3),village(B1,B2,B3),scenic-spot(C1,C2,C3)and field(D1,D2,D3)group.The filtered samples were performed by high-throughput sequencing and bioinformatics analysis was carried out to analyze the community structure of bacteria and archaea.[Result]There are significant differences in microbial diversity among samples,and the highest diversity and richness of bacteria and archaea appeared in the scenic-spot group.The interaction network of microbial species in springs was complex.The dominant bacterial phyla were Proteobacteria,Actinobacteria and Bacteroidetes,and Parvarchaeota,Woesearchaeota and Verrucomicrobia phylum were the dominant archaea.Under the influence of different degrees of disturbance,the specific dominant bacteria appeared in some springs,such as Helicobacteraceae,Verrucomicrobiaceae,Deferribacteres.[Conclusion]There were abundant bacteria and archaea in the springs of Wudalianchi volcanic area.The diversity,composition and abundance of bacteria and archaea in Wudalianchi volcanic area are different due to the disturbance of human activities.
