复方参及合剂对小鼠抗缺氧及疲劳的作用研究

摘  要:目的探讨复方参及合剂对小鼠抗缺氧及疲劳的作用。方法采用常压密闭缺氧实验、亚硝酸钠中毒实验检测小鼠抗缺氧能力,记录小鼠存活时间及小鼠脑、心肌组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)水平;采用负重游泳实验检测小鼠耐疲劳能力,记录小鼠力竭游泳时间,同时检测血清中三磷酸腺苷(ATP)、尿素氮(BUN)、肌糖原(MG)、肝糖原(Gly)、乳酸(LA)、乳酸脱氢酶(LDH)水平。3个实验均取KM小鼠50只,随机分为空白对照组(等体积生理盐水),阳性对照组(红景天胶囊0.3 g/kg),复方参及合剂低、中、高剂量组(简称低、中、高剂量组;1.0,2.0,3.0 g/kg),各组小鼠灌胃等体积生理盐水或相应药物(10 mL/kg),每日1次,连续14 d。结果与空白对照组比较,阳性对照组及低、中、高剂量组小鼠常压密闭缺氧条件下存活时间均显著延长(P<0.05);脑组织中,高剂量组SOD,低、中、高剂量组GSH-Px水平均显著升高,阳性对照组及低、中、高剂量组MDA水平均显著降低;心肌组织中,高剂量组SOD水平,阳性对照组及低、中、高剂量组GSH-Px水平均显著升高,低、高剂量组MDA水平均显著降低(P<0.05)。与空白对照组比较,阳性对照组及低、中、高剂量组小鼠亚硝酸钠中毒条件下存活时间均显著延长(P<0.05);脑组织中,低、中剂量组SOD,中、高剂量组GSH-Px水平均显著升高;心肌组织中,阳性对照组及中剂量组SOD,中剂量组GSH-Px水平均显著升高,阳性对照组及低、中、高剂量组MDA水平均显著降低(P<0.05)。与空白对照组比较,低、中、高剂量组小鼠负重游泳力竭时间均显著延长;血清中,高剂量组小鼠ATP,低、中、高剂量组LDH,中剂量组Gly水平均显著升高,阳性对照组及低、中、高剂量组BUN和LA水平均显著降低(P<0.05)。结论复方参及合剂可对小鼠发挥抗缺氧及疲劳作用。Objective To investigate the anti-hypoxia and anti-fatigue effects of Compound Shenji Mixture on mice.Methods The anti-hypoxia ability of mice was detected by the closed normobaric hypoxia test and sodium nitrite poisoning test,the survival time of mice was recorded,the superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-Px)levels in the brain and myocardial tissue of mice were detected.The anti-fatigue ability of mice was detected by the weight-loaded swimming test,the exhaustive swimming time of mice was recorded,and the adenosine triphosphate(ATP),urea nitrogen(BUN),muscle glycogen(MG),hepatic glycogen glycogen(Gly),lactate(LA),lactate dehydrogenase(LDH)levels in serum were detected.In the above three tests,50 KM mice were selected and randomly divided into the blank control group(equal volume of physiological saline),the positive control group(0.3 g/kg of Hongjingtian Capsules),the Compound Shenji Mixture low-,medium-and high-dose groups(referred to as the low-,medium-and high-dose groups,1.0,2.0,3.0 g/kg).The mice in each group were given an equal volume of physiological saline or corresponding drug(10 mL/kg)by gavage for 14 d once a day.Results Compared with those in the blank control group,the survival time in the positive control group,low-,medium-and high-dose groups in the closed normobaric hypoxia test significantly prolonged(P<0.05);the SOD level in brain tissue in the high-dose group,the GSH-Px level in brain tissue in the low-,medium-and high-dose groups significantly increased,the MDA level in brain tissue in the positive control group,low-,medium-and high-dose groups significantly decreased;the SOD level in myocardial tissue in the high-dose group,the GSH-Px level in myocardial tissue in the positive control group,low-,medium-and high-dose groups significantly increased,and the MDA level in myocardial tissue in the low-and high-dose groups significantly decreased(P<0.05).Compared with those in the blank control group,the survival time in the positive control group,low-,med
