摘 要:目的:利用孟德尔随机化方法探究肠道菌群与肾结石的相关性。方法:将MiBionGen联盟汇总的24个队列共18340例参与者的基因数据,196个肠道菌群亚群被纳入进行筛选,将与人类肠道菌群相关联并有统计学意义的单核苷酸多态性(SNP)作为工具变量。从英国生物样本库(UKB)中提取肾结石数据(ukb-b-8297)作为结局变量。利用逆方差加权法(IVW)和加权中位数法(WME)进行孟德尔随机化(MR)分析,绘制工具变量相关的肠道菌群与肾结石风险的森林图及散点图,探究肠道菌群与肾结石的相关性关系。结果:共筛选到12个与肠道菌群相关的SNP作为工具变量,IVW结果显示Coriobacteriaceae科与肾结石发病风险呈相关[OR=1.003;95%CI:(1.001,1.005);P=0.016],提示Coriobacteriaceae科丰度增加可促进肾结石的发生。结论:MR法分析显示,Coriobacteriaceae科与肾结石发病风险呈正相关。Objective With mendelian randomization method to explore the correlation between gut microbiota and kidney stones.Methods Using genetic data from a total of 18340 participants from 24 cohorts aggregated by the MiBionGen consortium,196 subpopulations of gut microbiota were included for screening,with statistically significant single nucleotide polymorphisms(SNPs)associated with human gut microbiota as instrumental variables.Extract kidney stones data(ukb-b-8297)from the UK Biological Sample Bank(UKB)as outcome variables.Using inverse variance weighted(IVW)and weighted median(WME)methods for Mendelian randomization(MR)analysis,draw forest and scatter plots of instrumental variable related gut microbiota and kidney stones risk,and explore the correlation between gut microbiota and kidney stones.Results A total of 12 SNPs related to the gut microbiota were screened as instrumental variables.The IVW results showed a correlation between the family Coriobacteriaceae and the risk of kidney stones[OR=1.003;95%CI:(1.001,1.005);P=0.016],suggesting that an increase in the abundance of Coriobacteriaceae may promote the occurrence of kidney stones.Conclusions MR analysis shows that Coriobacteriaceae family have a positive causal effect on the risk of kidney stones.
