摘 要:目的:阐明肤黄止痒汤对尿毒症皮肤瘙痒的疗效及其作用机制。方法:将40只小鼠随机分为5组(Sham、Model、FHZYT、BCTC、FHZYT+BCTC),每组8只,干预完成后计数小鼠搔抓次数。指标检测包括免疫组化染色检测皮肤中TNF-α、IL-1β、Substance P和IL-31的表达。WB检测皮肤中Substance P、TNF-α、IL-1β、IL-31、TRPV1、ERK1/2和p-ERK1/2的蛋白表达。结果:1.Model组小鼠肌酐、尿素氮水平升高,瘙痒次数增加(P<0.01)。2.Model组中TNF-α、IL-1β的表达上调,FHZYT、BCTC、FHZYT+BCTC组TNF-α表达则降低(P<0.01)。3.与Model组相比,FHZYT、BCTC、FHZYT+BCTC组瘙痒次数相对减少,皮肤中P物质、IL-31、TRPV1、ERK1/2及p-ERK1/2的表达水平降低(P<0.01)。结论:肤黄止痒汤可有效减少尿毒症小鼠皮肤瘙痒,其机制可能是通过抑制IL-31/TR⁃PV1/ERK1/2信号轴,改善皮肤炎症状态并发挥止痒作用。Objective:To elucidate the efficacy of Fu Huang Zhi Yang Tang on Uremic Pruritus and its mechanism of action.Methods:40mice were randomly divided into 5groups(Sham,Model,FHZYT,BCTC,FHZYT+BCTC),8mice in each group,and the number of scratching was counted after the intervention was completed.Indicator tests included immunohistochemical staining to detect the expression of TNF-α,IL-1β,Substance P and IL-31in the skin.WB to detect the protein expression of Substance P,TNF-α,IL-1β,IL-31,TRPV1,ERK1/2 and p-ERK1/2 in the skin.Results:1.Mice in the Model group had increased levels of creatinine and urea nitrogen,and the number of itching increased.2The expression of TNF-αand IL-1βwas up-regulated in the Model group,whereas the expression of TNF-αwas decreased in the FHZYT,BCTC,and FHZYT+BCTC groups.3Compared with the Model group,the number of itching in the FHZYT,BCTC,and FHZYT+BCTC groups was relatively The expression levels of substance P,IL-31,TRPV1,ERK1/2 and p-ERK1/2 in the skin were reduced.Conclusion:Fu Huang Zhi Yang Tang can effectively reduce itching in uremic mice,and its mechanism may be through the inhibition of IL-31/TRPV1/ERK1/2 signalling axis,which improves the inflammatory state of the skin and exerts the effect of itching relief.
