少腹逐瘀汤减少E_(2)、PGE_(2)合成抑制内异症模型大鼠异位病灶的机制研究  被引量:2

摘  要:目的:基于大鼠模型探讨少腹逐瘀汤治疗内异症的可能机制。方法:以自体子宫移植法建立SD大鼠内异症模型,造模成功的大鼠随机分为模型组、来曲唑组、少腹逐瘀组3组,每组12只。模型组给予生理盐水、药物治疗组分别给予相应的药物干预4周。记录给药前后异位病灶体积。ELISA法检测各组循环中性激素水平、异位病灶局部E_(2)、E1、PGE_(2)水平;免疫组化及Western Blot法检测异位病灶中COX-2、StAR、aromatase、ERα和ERβ表达。结果:来曲唑和少腹逐瘀汤都能够缩小异位病灶体积。来曲唑能够降低内异症大鼠循环中及异位病灶局部E_(2)、E1水平,上调循环中FSH、LH水平;少腹逐瘀汤能够降低内异症大鼠异位病灶局部E_(2)、E1水平,上调循环中FSH水平;两种药物都可以降低异位病灶局部COX-2、PGE_(2)水平,对StAR、aromatase表达无影响;来曲唑能够上调异位病灶局部ERα表达但对ERβ无影响,少腹逐瘀汤不仅能够上调ERα表达还能够下调ERβ表达。结论:少腹逐瘀汤能够通过降低COX-2、PGE_(2)表达起到减少异位病灶局部E_(2)合成的作用,同时调整ERα/ERβ比值,以上可能是其治疗内异症的机制。Objctive The aim of this study was to determine the mechanism of Shafu Zhuyu Decoction(SFZYD)in a rat endometriosis model.Methods Autologous uterine transplantation was performed in Sprague Dawley(SD)rats,and those with successful ectopic implants were randomly divided into model group,letrozole group and SFZY group,and were treated with saline placebo,letrozole or SFZYD for a period of 4weeks.The implant volume was recorded before and after medication,and serum sex hormone levels as well as the levels of estrogen and prostaglandin(PG)E_(2)in the ectopic lesions were determined by ELISA.The expression of COX-2,StAR,aromatase,ERαand ERβin the lesions was detected by immunohistochemistry and Western blotting.Results Both letrozole and SFZYD reduced the size of endometriosis lesions.Letrozole decreased E_(2)and E1 levels in both serum and lesions,increased FSH and LH levels in serum;SFZYD only targeted E_(2)and E1 levels in the lesions and increased FSH in serum.Both drugs reduced local COX-2expression and inhibited PGE_(2)production without affecting StAR and aromatase expression.In addition,letrozole upregulated ERαwith no significant impact on ERβ,whereas SFZYD not only up-regulated ERαbut also down-regulated ERβ.Conclusion The mechanism of Shaofu Zhuyu Decoction on endometriosis mice may be realized by reducing E_(2)synthesis in the ectopic lesions by lowering COX-2expression and inhibiting PGE_(2)production,along with balancing ERα/ERβexpression ratio.
