摘 要:目的:基于TOLL样受体4(TLR4)介导的髓样分化因子88(MyD88)/核因子-κB(NF-κB)通路探讨通平脂肝方对高脂饲料诱导的非酒精性脂肪肝(non-alcoholicfattyliverdisease, NAFLD)模型小鼠的肝脏保护作用及抑制炎症的相关机制。方法:选用C57BL/6J雄性小鼠60只,随机分为正常、模型、盐酸吡格列酮和通平脂肝方低、中、高剂量组,各10只。给药8 w后取材。结果:与正常组相比,模型组体质量、血脂、肝功能、IL-6、TNF-α及TLR4、MyD88、Ikkβ、NF-κB蛋白和TLR4、MyD88、Ikkβ的mRNA表达水平均上升(P<0.05),通平脂肝方中、高剂量组显著改善上述指标(P<0.05)。模型组肝细胞大量脂肪蓄积,气球样变性,细胞质受到球形脂滴占位性挤压以及炎症细胞浸润,各用药组肝组织病理形态得到改善,球形脂滴及炎性细胞浸润减少。结论:通平脂肝方可调节高脂饮食诱导的NAFLD小鼠的血脂水平,改善肝功能,减少肝脏脂质堆积,减轻脂肪肝及肝损伤,减轻炎症反应。其抗炎机制可能与其激活TLR4介导的MyD88/NF-κB信号通路,抑制脂质合成及炎症因子的表达有关。Objective:To explore the protective effect of Tongping Zhigan Formula on high-fat-diet-induced non-alcoholic fatty liv-er disease(NAFLD)in model mice and its related mechanisms of inhibiting inflammation,based on myeloid differentiation factor 88(MyD88)/nuclear factor-κB(NF-κB)pathway mediated by Toll-like receptor 4(TLR4)Methods:C57BL/6J male mice were ran-domly divided into normal group,model group,pioglitazone hydrochloride group,and low-,medium-,and high-dose groups of Tong-ping Zhigan Formula,with 10 mice in each group Samples were collected after 8 weeks of administration Results:Compared with the normal group,the indicators in model group,including body mass,blood lipids,liver function,IL-6,TNF-α,and TLR4,MyD88,Ikkβ,NF-κB protein,and mRNA expressions of TLR4,MyD88 and Ikkβ,increased significantly(P<005);The above indicators of the midium-and high-dose groups of Tongping Zhigan Formula significantly improved(P<005)A large amount of fat accumulated in the liver cells in the model group,with ballooning degeneration;The cytoplasm was compressed by spherical lipid droplets and infil-trated by inflammatory cells;The pathological morphology of liver tissue in each medication group was improved,and the infiltration of spherical lipid droplets and inflammatory cells were reduced Conclusion:Tongping Zhigan Formula can regulate blood lipid levels in NAFLD mice induced by high-fat diet,improve liver function,reduce liver lipid accumulation,alleviate fatty liver and liver damage,and alleviate inflammation Its anti-inflammatory mechanism may be related to its activating TLR4-mediated MyD88/NF-κB signaling pathway,inhibiting lipid synthesis and the expression of inflammatory factors.
