摘 要:为了探究Nesfatin-1对肥胖大鼠慢性炎症的影响,试验以健康SPF级雄性SD大鼠为对照组,通过高脂饮食诱导肥胖大鼠模型,并将造模成功的28只肥胖大鼠随机分为模型对照组(生理盐水)、Nesfatin-1低剂量(1μg/kg)组、Nesfatin-1中剂量(10μg/kg)组和Nesfatin-1高剂量(100μg/kg)组,每组7只。Nesfatin-1低、中、高剂量组大鼠连续尾静脉注射不同浓度Nesfatin-1,5周后测量体重、体长,计算Lee’s指数,取肝脏并进行H.E.染色观察病理变化;采用ELISA法检测各组大鼠血清肿瘤坏死因子-α(TNF-α)和白细胞介素-10(IL-10)含量的变化;运用qPCR法检测各组大鼠肝脏miR-155-5p和miR-146a-5p的相对表达量,以评估Nesfatin-1对慢性炎症的调节能力。结果表明:模型对照组大鼠的体重和Lee’s指数均极显著高于对照组(P<0.01)。不同浓度的Nesfatin-1均极显著抑制肥胖大鼠的体重增长(P<0.01),并且对肝脏脂肪变性有改善作用。与模型对照组比较,Nesfatin-1低剂量组炎症因子TNF-α含量显著降低(P<0.05),Nesfatin-1低、中、高剂量组抗炎细胞因子IL-10含量均显著升高(P<0.05);Nesfatin-1高剂量组miR-155-5p相对表达量显著降低(P<0.05),Nesfatin-1低剂量组miR-146a-5p相对表达量显著升高(P<0.05),Nesfatin-1高剂量组miR-146a-5p相对表达量极显著升高(P<0.01)。说明Nesfatin-1能够通过抑制肥胖大鼠体重增长、肝脏脂肪变性调节肥胖大鼠血清炎症因子水平和肝脏中miR-146a-5p、miR-155-5p的相对表达量。In order to investigate the effects of Nesfatin-1 on chronic inflammation in obese rats,in the experiment,healthy SPF grade male SD rats were used as the control group;the obese rat model was induced by high-fat diet;twenty-eight obese rats of successful modeling were randomly divided into the model control group(saline),the Nesfatin-1 low dose(1μg/kg)group,the Nesfatin-1 medium dose(10μg/kg)group,and the Nesfatin-1 high dose(100μg/kg)group with 7 rats in each group.The rats in the Nesfatin-1 low,medium and high dose groups were injected continuously into the tail vein with different concentrations of Nesfatin-1.After 5 weeks,body weight and length were measured,and Lee's index was calculated;livers were taken and H.E.staining was used to observe the pathological changes.ELISA was used to detect the changes of tumor necrosis factor-α(TNF-α)and interleukin-10(IL-10)in the serum of rats in each group.The relative expression of miR-155-5p and miR-146a-5p in the liver of rats in each group was detected by using qPCR to assess the regulatory ability of Nesfatin-1 on chronic inflammation.The results showed that the body weight and Lee's index of rats in the model control group were both extremely significantly(P<0.01)higher than those in the control group.Different concentrations of Nesfatin-1 highly significantly inhibited the weight gain of obese rats(P<0.01)and had an ameliorating effect on hepatic steatosis.Compared with the model control group,the level of inflammatory factor TNF-αwas significantly reduced in the Nesfatin-1 low-dose group(P<0.05);and the levels of anti-inflammatory cytokine IL-10 were significantly elevated in the Nesfatin-1 low-,medium-and high-dose groups(P<0.05);the relative expression of miR-155-5p in the Nesfatin-1 high-dose group was significantly lower(P<0.05);the relative expression of miR-146a in the Nesfatin-1 low-dose group was significantly higher(P<0.05);the relative expression of miR-146a-5p in the Nesfatin-1 high-dose group was extremely significant(P<0.01).The results indicat
