摘  要:为了探究妊娠早期饲喂N-氨甲酰谷氨酸(N-Carbamylglutamate,NCG)提高山羊产羔数的繁殖机制,试验采用实时荧光定量PCR方法检测了ITGB8、IGFBP3基因在空白组和NCG组(0.15%NCG)卵巢、子宫、输卵管、垂体、下丘脑等性腺轴组织中的相对表达量。结果表明:ITGB8和IGFBP3基因在贵州白山羊5个组织中均有不同程度表达。相同组别不同组织比较,ITGB8基因在空白组的相对表达量为下丘脑>垂体>输卵管>子宫>卵巢,在NCG组的相对表达量为垂体>子宫>下丘脑>输卵管>卵巢;IGFBP3基因在空白组和NCG组中的表达趋势均为子宫>卵巢>输卵管>下丘脑>垂体。相同组织不同组间比较,ITGB8基因在NCG组垂体、卵巢、输卵管、子宫组织中的相对表达量极显著低于空白组(P<0.01),在下丘脑中两组间差异不显著(P>0.05);IGFBP3基因在NCG组垂体、卵巢、输卵管、子宫组织中的相对表达量均极显著低于空白组(P<0.01),在下丘脑中两组间差异不显著(P>0.05)。说明给山羊饲喂NCG能提高ITGB8基因和降低IGFBP3基因的相对表达量,NCG可能通过调控ITGB8和IGFBP3基因的表达影响山羊繁殖性能。In order to investigate the reproductive mechanism of feeding N-carbamylglutamate(NCG)in early gestation to increase lambing number in goats,in the experiment,the relative expression of ITGB8 and IGFBP3 genes in the gonadal tissues of ovary,uterus,oviduct,pituitary gland and hypothalamus in the blank group and NCG group(0.15%NCG)was detected by real-time fluorescence quantitative PCR.The results showed that ITGB8 and IGFBP3 genes were expressed in different degrees in five tissues of Guizhou White goats.Comparing different tissues in the same group,the relative expression of ITGB8 gene in the blank group was hypothalamus>pituitary>oviduct>uterus>ovary;while in the NCG group,the relative expression was pituitary>uterus>hypothalamus>oviduct>ovary.The expression trend of IGFBP3 gene in the blank group and the NCG group was both uterus>ovary>oviduct>hypothalamus>pituitary.Comparing between different groups of the same tissue,the relative expression of ITGB8 gene in the pituitary,ovary,fallopian tube and uterus tissues of the NCG group was extremely significantly lower than that of the blank group(P<0.01),and the difference between the two groups in the hypothalamus was not significant(P>0.05).The relative expression of IGFBP3 gene in the pituitary,ovary,oviduct and uterus tissues of the NCG group was extremely significantly lower than that of the blank group(P<0.01),and the difference between the two groups in the hypothalamus was not significant(P>0.05).The results indicated that feeding NCG to goats could increase the relative expression of ITGB8 gene and decrease IGFBP3 gene,and NCG might affect the reproductive performance of goats by regulating the expression of ITGB8 and IGFBP3 genes.
