摘 要:目的:探讨龟鹿二仙胶对大鼠绝经后骨质疏松性骨折(postmenopausal osteoporotic fractures,PMOPF)愈合的影响。方法:从34只SD大鼠中随机选取11只作为对照组,仅切除卵巢旁脂肪组织,其余大鼠行双侧卵巢切除术建立骨质疏松模型。12周后,所有大鼠行右侧股骨骨折合并髓内针固定术。将造模成功的PMOPF大鼠随机分为模型组、龟鹿二仙胶组(3.6 g·kg^(-1))和阿仑膦酸钠组(3.6 g·kg^(-1)),每组6只。龟鹿二仙胶组和阿仑膦酸钠组给予相应药物灌胃,对照组和模型组给予等剂量生理盐水灌胃,持续8周。采用HE染色、番红固绿染色观察骨折处病理形态;三点弯曲法检测骨折处最大载荷;ELISA法检测血清骨钙素(bone gamma carboxy-glutamic acid containing proteins,BGP)、骨碱性磷酸酶(bone alkaline phosphatase,BALP)、Ⅰ型胶原交联C-末端肽(type I collagen carboxy-terminal peptide,CTX-1)含量;RT-qPCR和Western blot检测骨折处骨痂BGP、BALP、骨细胞特异性转录因子(Osterix)蛋白和基因表达水平。结果:HE染色结果显示,对照组大鼠骨折处初步形成未成熟骨组织,构成编织骨;模型组大鼠骨折处仅见少量未成熟骨组织;与模型组比较,龟鹿二仙胶组、阿仑膦酸钠组大鼠骨折处骨组织更成熟,含有大量编织骨。番红固绿染色结果显示,对照组大鼠骨折处以钙化骨痂为主,含有残存软骨组织;模型组大鼠骨折处含有大量未完成钙化的软骨组织;龟鹿二仙胶组、阿仑膦酸钠组大鼠骨折处含有残存软骨组织,已形成较多钙化骨痂。与对照组比较,模型组大鼠骨折处最大载荷降低(P<0.01);与模型组比较,龟鹿二仙胶组、阿仑膦酸钠组大鼠骨折处最大载荷升高(P<0.01)。与对照组比较,模型组大鼠血清BGP、BALP含量减少且CTX-1含量增加(P<0.01);与模型组比较,龟鹿二仙胶组、阿仑膦酸钠组大鼠血清BGP、BALP含量增加且CTX-1含量减少(P<0.05)。与对照组比较,模型组大鼠Objective:To investigate the effect of Guilu Erxian Gum on the healing of postmenopausal osteoporotic fracture(PMOPF)in rats.Methods:Eleven SD rats were randomly selected from 34 SD rats as the control group,with only the para-ovarian adipose tissue was removed,while the rest rats underwent bilateral oophorectomy to establish the osteoporosis model.After 12 weeks,all rats underwent right femoral fracture with intramedullary nailing fixation.The PMOPF rats with successful modeling were randomly divided into a model group,a Guilu Erxian Gum group(3.6 g·kg^(-1))and an alendronate sodium group(3.6 g·kg^(-1)),with 6 rats in each group.The Guilu Erxian Gum group and alendronate sodium group were given the corresponding drugs by gavage,while the control group and model group were given the same dose of normal saline by gavage for 8 weeks.HE staining and Safranin fast green staining were used to observe the pathological morphology of the fracture.The maximum load at the fracture site was detected by"three-point bending"method.ELISA was used to detect serum levels of bone gamma carboxy-glutamic acid containing proteins(BGP)and bone alkaline phosphatase(BALP)and type I collagen carboxy-terminal peptide(CTX-1).RT-qPCR and Western blot were used to detect the gene and protein expression levels of BGP,BALP,and Osterix in callus at the fracture site.Results:HE staining results showed that immature bone tissue was initially formed at the fracture site in the control group,which constituted woven bone.In the model group,only a small amount of immature bone tissue was observed at the fracture site.Compared with the model group,the bone tissue in the Guilu Erxian Gum group and alendronate sodium group was more mature and contained a large amount of woven bone.Safranin fast green staining results showed that the fracture area of the control group was dominated by calcified callus containing residual cartilage tissue.The fracture site of rats in the model group contained a large number of incompletely calcified cartilage tissue.Th
