摘 要:目的基于网络药理学探讨补肾与健脾中药配伍治疗骨质疏松症(OP)的潜在作用机制。方法阅读文献,选用出现频率较高的补肾药与健脾药,从中药系统药理学数据库与分析平台(TCMSP)中筛选出其有效成分,并搜集有效靶点。结果补肾药共筛选出有效成分60个、预测靶点199个,健脾药筛选出有效成分40个、预测靶点195个;检索并筛选出OP疾病靶点1531个,预测靶点与OP共有交集72个;蛋白质互作网络(PPI)分析提示核心靶点为肿瘤蛋白p53基因(TP53)等;其中糖尿病并发症中的高级糖基化终末产物-受体(AGE-RAGE)信号通路、流体剪应力与动脉粥样硬化等可能为治疗OP的关键通路。结论补肾药与健脾药可通过多成分、多靶点、多通路作用于OP的治疗。Objective To explore the potential mechanism of the combination of traditional Chinese medicines for nourishing kidney and strengthening spleen in the treatment of osteoporosis(OP)based on network pharmacology.Methods After reading the literature,nourishing kidney and strengthening spleen drugs with high frequency were selected,and their effective components were screened from the TCM Systematic Pharmacology database and analysis platform(TCMSP),and effective targets were collected.Results A total of 66 effective components and 199 predicted targets were selected for kidney tonifying drugs,and 44 effective components and 195 predicted targets were selected for spleen tonifying drugs.A total 1531 disease targets of OP were retrieved and screened,and 72 tar⁃gets were predicted to intersect with OP.Protein interaction network(PPI)analysis suggested that the core targets were tumor protein p53 gene(TP53),MYC,etc.In the gene ontology(GO)analysis,1970 biological processes,44 cell components and 90 molecular func⁃tions were obtained.A total of 157 results were obtained from the Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis,among which advanced glycation end-product receptor(AGE-RAGE)signaling pathways in diabetes complications,fluid shear stress,and atherosclerosis may be key pathways in the treatment of OP.Conclusion The nourishing kidney and strengthening spleen medicine can act on the treatment of OP through multi-component,multi-target and multi-pathway.
