补肾活血汤经miR-135b改善内异症大鼠子宫内膜容受性的机制研究

摘　　要：目的观察补肾活血汤对子宫内膜异位症(EMs)模型大鼠种植窗期子宫内膜miR-135b和HOXA10 mRNA的影响,探讨子宫内膜容受性(ER)的影响机制及补肾活血汤对EMs大鼠ER的改善机制。方法建立EMs的大鼠模型,随机分为空白正常组、假手术组、模型组、西药组、中药组,采用RT-PCR法检测各组大鼠子宫内膜miR-135b、HOXA10 mRNA的表达。结果空白正常组和假手术组种植窗口期子宫内膜miR-135b表达量显著低于模型组、中药组和西药组,中药组显著低于模型组和西药组(P<0.001);空白正常组和假手术组的HOXA10 mRNA表达量显著高于模型组、中药组和西药组,而中药组和西药组显著高于模型组(P<0.001)。结论补肾活血汤可通过下调小鼠子宫内膜miR-135b,从而上调HOXA10 mRNA的表达,改善ER。Objective To investigate the mechanism of endometrial receptivity(ER)and the improvement mechanism of Bushen Huoxue decoction on ER in endometriosis(EMs)model rats,as well as the influence of Bushen Huoxue decoction on miR-135b and HOXA10 mRNA in the endometrium during the implantation window time.Methods Create a rat model of EMs and randomly assign each group to one of the following:blank normal,sham operation,model,Western,and Chinese medicine.The expression of miR-135b and HOXA10mRNA in each group of rats was determined using real-time PCR.Results The expression level of miR-135b in the endometrium of the blank normal group and sham surgery group during the implantation window period was significantly lower than that of the model group,Chinese medicine group,and Western medicine group,while the Chinese medicine group was signifi⁃cantly lower than that of the model group and Western medicine group(P<0.001).The HOXA10 mRNA expression levels in the blank normal group and sham surgery group were significantly higher than those in the model group,Chinese medicine group,and Western medicine group.The expression levels of HOXA10 mRNA in the Chinese medicine group and the Western medicine group were sig⁃nificantly higher than those in the model group(P<0.001).Conclusion Bushen Huoxue decoction can can up-regulate the expression of HOXA10 mRNA and improve ER by down-regulating miR-135b in mouse endometrium.
