摘  要:磷酸肌醇(PI)是多种细胞功能的重要调节剂,含有SAC(Suppressor of actin)结构域的磷酸肌醇磷酸酶参与PI合成,称为SAC磷酸酶,目前SAC磷酸酶在木本植物中研究较少,为了探究SAC磷酸酶对黄梁木的生长发育和抗逆性的影响,本研究利用生物信息学方法鉴定得到10个黄梁木SAC基因,并对其编码的蛋白进行系统进化和表达模式分析。结果显示,NcSACs蛋白序列长度为508~1734 aa,NcSACs蛋白有4个显酸性和6个显碱性,且均为亲水性蛋白,其中包含4个稳定蛋白,NcSACs蛋白具有多个潜在磷酸化位点,可通过磷酸化位点调节NcSACs酶活性。10个NcSACs蛋白都具有保守的SAC结构域,且NcSACs蛋白可根据C端序列的差异性被分为Ⅰ、Ⅱ、Ⅲ3个亚型。系统进化分析也显示NcSACs分为Ⅰ、Ⅱ、Ⅲ3种亚型,且NcSAC8和NcSAC9与At SAC6和AtSAC7亲缘关系较近,推测具有相近的功能,在植物根系伸长以及盐胁迫抗逆方面具有重要意义。通过转录组测序技术,对NcSACs进行表达模式分析,结果表明,NcSACs家族基因的表达有组织特异性,其中NcSAC4在果实、根系和老叶中有较高的表达量。本研究为深入研究NcSACs基因的功能以及探索SAC磷酸酶对林木生长发育及抗逆的影响提供了理论基础。Phosphoinositide(PI)is an important regulator of a variety of cell functions.The Suppressor of Actin(SAC)domain-containing proteins are a class of phosphoinositide phosphatase involved in the synthesis of PIs,called SAC phosphatase,but there is little research on SAC phosphatase on trees.In order to explore the influence of SAC phosphatase on the growth and stress resistance of Neolamarckia cadamba,this study identified 10 SAC genes and analyzed proteins for evolutionary and expression pattern analysis in Neolamarckia cadamba.The results show that the sequence length of NcSAC proteins is 508~1734 aa.NcSAC proteins have 4 acidic and 6 basic,and they are all hydrophilic proteins,and 4 of them are stable proteins.NcSAC proteins have multiple potential phosphorylation sites.In particular,the enzyme activity of NcSACs can be regulated by phosphorylation sites.10 NcSAC proteins have the conserved SAC domain,and NcSAC proteins can be divided into three subtypes:Ⅰ,Ⅱ and Ⅲ,according to the differences in C-terminal sequences.The phylogenetic analysis of NcSACs also shows that NcSACs gene family can be divided into three subtypes:Ⅰ,Ⅱ and Ⅲ,and NcSAC8 and NcSAC9 are closely related to AtSAC6 and AtSAC7,which are speculated to have similar functions.It is of great significance in terms of growth and resistance to salt stress.The expression pattern analysis of NcSACs showed that the NcSACs family genes are differentially expressed and may play dominant roles in particular organs,and NcSAC4 has higher expression amount in fruits,roots and old leaves.This study provides a theoretical basis for the in-depth study of the function of the NcSACs and exploring the effect of SAC phosphatase on the growth and development of trees.
