摘  要:端粒酶(Telomerase)主要通过合成端粒DNA使得端粒延长,以维持端粒的完整性,而端粒酶逆转录酶(telomerase reverse transcriptase,TERT)是端粒酶的核心组分。目前关于多年生草本植物毛竹PeTERT基因相关研究还未见报道。本研究对PeTERT基因进行了生物信息学分析,采用RT-qPCR技术检测毛竹PeTERT在不同组织及外源激素和非生物胁迫处理后的表达变化。结果表明:在毛竹基因组中鉴定到PeTERT基因序列,CDS序列全长3789 bp,PeTERT含有1262个氨基酸,等电点为9.45,相对分子质量为143.74 kD,含有TRBD和RT保守结构域,二级和三级结构具有TERT的基本特征;PeTERT启动子序列包含多个响应激素和非生物胁迫元件;PeTERT在细胞分裂能力强的毛竹愈伤组织中表达量为幼苗根、茎、叶的2.5~5倍;外源激素ABA、MeJA、GA_(3)、SA均可不同程度地影响毛竹幼苗PeTERT的表达,同时PeTERT可响应低温(4℃)、高温(42℃)、高盐(200 mmol/L)、干旱(20%PEG-6000)等多种逆境条件,表明毛竹PeTERT具有非端粒功能,研究结果为探讨该基因的生物学功能提供了重要参考。Telomerase is essential for maintaining telomere integrity.Telomerase reverse transcriptase(TERT)is the core component of telomerase,which lengthens telomeres mainly by synthesizing telomere DNA to maintain telomere integrity.At present,there have been no reports on the PeTERT gene of Phyllostachys edulis.In this study,bioinformatics analysis was carried out on PeTERT,and RT-qPCR was used to detect the PeTERT expression patterns in leaf,stem,root and callus and seedlings treated with exogenous hormones and abiotic stress.The results showed that the PeTERT gene sequence was identified in Phyllostachys edulis genome.The CDS sequence is 3789 bp,PeTERT contains 1262 amino acids,the isoelectric point is 9.45,the relative molecular mass is 143.74 kD,showing the basic characteristics of the secondary and tertiary structures of TERT with TRBD and RT conservative domain structure.The PeTERT promoter sequence contains multiple hormone and abiotic stress response elements.RT-qPCR detected the expression of PeTERT in Phyllostachys edulis,and the expression level in callus with strong cell division ability was 2.5~5 times higher than that in roots,stems and leaves of seedlings.The exogenous hormones ABA,MeJA,GA_(3) and SA could affect the expression of PeTERT in seedlings,and PetERT could also respond to stress conditions such as low temperature(4℃),high temperature(42℃),high salt(200 mmol/L),and drought(20%PEG-6000),indicating that PeTERT may exert non-telomere function.This research provides important reference for studying the biological function of PeTERT gene.
