摘  要:芒果细菌性角斑病是芒果生产上的重要细菌病害之一,为揭示芒果品种对细菌性角斑病的抗性差异,本研究通过全基因组重测序技术对高抗细菌性角斑病品种‘热农1号’和高感细菌性角斑病品种‘凯特’进行测序,通过与参考基因组‘红象牙’进行比对,进行数据统计与质量检测、序列比对及多态性SNP位点分析。结果表明,在测序深度10X下,‘凯特’和‘热农1号’分别获得5.93 Gb和4.55 Gb的数据量,过滤后的Q30达到92.4%和92.3%。将获得的原始数据与参考基因组对比,‘凯特’和‘热农1号’比对上的reads数目分别是31108817和30217182。‘凯特’和‘热农1号’分别检测到3530706和3325884个SNP位点,产生非同义突变的SNP位点数分别为133283和127148个,杂合度为0.43%和0.36%,转换和颠换的比值为2.24和2.25。基于抗病品种‘热农1号’和感病品种‘凯特’全基因组重测序数据比较的结果,获得到了3258857个差异SNP位点和33161条基因信息,其中有937个基因参与抗病蛋白的调控。本研究结果可以在一定程度上反映抗感细菌性角斑病的芒果品种在基因组水平抗性差异,可为下一步细菌性角斑病杂交群体材料的抗性评价和抗病特异性分子标记开发提供基础数据,同时为后期芒果抗病优质品种的创制和选育提供理论依据。Mango bacterial black spot disease is one of the important bacterial diseases in mango production.In order to reveal the difference in resistance of mango varieties to bacterial black spot disease,this study used whole-genome resequencing technology to identify highly resistant bacterial black spot disease varieties‘Renong No.1’and the highly susceptible bacterial black spot disease variety‘Keitt’were sequenced,and compared with the reference genome‘Red Ivory’,data statistics and quality detection,sequence comparison and polymorphic SNP locus analysis were performed.The results showed that at a sequencing depth of 10X,‘Keitt’and‘Renong No.1’obtained 5.93 Gb and 4.55 Gb data respectively,and the filtered Q30 reached 92.4% and 92.3%.Comparing the obtained original data with the reference genome,the number of reads in the comparison of‘Keitt’and‘Renong No.1’are 31108817 and 30217182,respectively.3530706 and 3325884 SNP loci were detected in‘Keitt’and‘Renong No.1’,respectively.The number of SNP loci that produced non-synonymous mutations were 133283 and 127148,respectively,and the heterozygosity was 0.43% and 0.36%,the ratio of conversion and transversion is 2.24 and 2.25.In the annotation analysis of gene function,3258857 differential SNP loci and 33161 pieces of gene information were obtained,of which 937 genes are involved in the regulation of disease-resistant proteins.The results of this study can reflect to a certain extent the differences in genomic resistance of mango varieties that are resistant to bacterial black spot disease,and can be used for the next step in the resistance evaluation of bacterial black spot disease hybrid population materials and the development of specific molecular markers for disease resistance,and provide theoretical basis for the creation and breeding of high-quality mango varieties resistant to disease in the later stage.
