人工感染PRRSV仔猪心脏的临床表现和病理变化

摘　　要：【目的】了解PRRSV感染对仔猪心脏的损伤特点。【方法】以PRRSV JXA1株感染约5周龄长白仔猪,出现临床症状后,使用听诊器检查人工感染组和正常对照组仔猪的心音变化,发病第3天,剖检并取左心室壁组织,制作石蜡切片,HE染色观察病理变化,免疫组织化学染色检测PRRSV N蛋白分布。【结果】心脏听诊结果显示,感染仔猪第二心音显著性增强,部分仔猪心动过速或心律不齐,偶有轻微心包拍水音;剖检结果显示,感染仔猪心包膜略增厚,心包腔少量积液,心脏横径略大,质地较软,心肌有出血点;HE染色结果显示,感染仔猪心肌组织出现淤血、淋巴细胞浸润、颗粒变性等病理性变化;免疫组织化学染色结果显示,PRRSV N蛋白在心肌组织分布广泛,不仅分布于心肌细胞,也分布于血管内皮细胞。【结论】PRRSV JXA1株人工感染第3天,即引起仔猪心脏功能变化和心肌组织病变,这表明在该病毒病的治疗中应尽早加强对心脏功能的保护。[Objective]To comprehend the heart damage of piglets caused by PRRSV infection.[Methods]Landrace piglets about 5 weeks old were artificially infected with PRRSV JXA1 strain.After the onset of obvious clinical symptoms,the heart sounds of piglets were diagnosed by a stethoscope.At three days post infection,piglets were autopsied and the ventricular parietal tissues were sampled to prepare the paraffin sections.The pathological changes of piglet hearts were detected by the Hematein-Eosin staining,and the PRRSV N protein were determined by immunohistochemical staining.[Results]Cardiac auscultation showed that the second heart sound of infected piglets was significantly enhanced,and there were tachycardia and arrhythmia and occasional pericardial patting sounds in some infected piglets.The cardiac autopsy showed that the slightly thickened pericardium,small amount of fluid in pericardial space,the slightly larger transverse cardiac diameter,the softer tissues,and some myocardial hemorrhagic spots in infected piglets.HE staining indicated myocardial tissue congestion,lymphocyte infiltration and granular degeneration,etc.Immunohistochemical staining indicated that PRRSV N protein was widely detected in myocardial tissue,not only in myocardial cells,but also in vascular endothelial cells.[Conclusion]Three days post infection with PRRSV JXA1 strain,cardiac function changes and pathological changes of myocardial tissues were directly caused,which suggests the cardiac function protection should be strengthened as soon as possible in the treatment of PRRSV-infected piglets.
