摘 要:为了解2021—2022年我国禽类养殖场中Ⅰ群禽腺病毒(GroupⅠfowladenovirus,FAdVⅠ)分子流行情况,试验通过病毒分离、PCR鉴定、鸡胚致病性试验、Hexon基因测序等方法对采集自2021—2022年我国15个地区共计250份疑似感染禽腺病毒的组织病料进行病原分析。结果显示:共分离出43株FAdVⅠ;Hexon基因扩增测序分析表明,其中36株与FAdV-C4型参考株核苷酸序列相似性在99.8%~100%,2株与FAdV-E8b型参考株核苷酸序列相似性达到93.8%~94.2%,1株与FAdV-A1型参考株核苷酸序列相似性达到99.5%,4株与FAdV-D11型参考株核苷酸序列相似性达到95.4%;FAdVⅠ感染在夏秋季节为高发期,阳性检出率较高的时间主要集中于4—10月。研究表明,我国15个主要养禽地区禽群中FAdVⅠ流行情况较为复杂,存在交叉感染和病毒基因重组风险,需要进一步调查其流行病学特征及研究其致病性。To investigate the molecular prevalence of GroupⅠavian adenovirus(FAdVⅠ)in poultry farms in China from 2021 to 2022,the virus isolation,PCR,chicken embryo pathogenicity test and Hexon gene sequencing were used to analyze the 250 suspected infected poultry adenovirus tissues collected from 15 regions from 2021 to 2022.The findings revealed that there were 43 isolates of FAdVⅠ.The Hexon genetic similarities between the 36 strains of FAdV-C and the FAdV-C4 reference strain were found to be remarkably high with a nucleotide sequence identity ranging from 99.8%to 100%.Similarly,2 strains of FAdV-E and the FAdV-E8b reference strain exhibited a nucleotide sequence identity of 93.8%to 94.2%.Furthermore,1 strain of FAdV-A and the FAdV-A1 reference strain shared a nucleotide sequence identity of 99.5%,while the 4 strains of FAdV-D and the FAdV-D11 reference strain displayed a nucleotide sequence identity of 95.4%.The epidemic periods primarily occurred during summer and autumn,with the highest incidence was from April to October.The results showed that prevalence and distribution of FAdVⅠserotype exhibited significant variations across the 15 regions in China,which meaned there were risks of cross infection and viral gene recombination,and it was imperative to further investigation of epidemiological characteristics and pathogenicity of FAdVⅠin China.
