摘 要:目的:利用网络药理学分析川牛膝治疗高血压的作用机制。方法:应用中药系统药理学数据库及分析平台(TCMSP)筛选出川牛膝的活性成分及其作用靶点,采用OMIM数据库,Drugbank数据库,TTD数据库,CTD数据库和DISGENT数据库检索与高血压相关的靶点基因。将川牛膝及高血压的共同靶点导入STRING数据库进行蛋白质-蛋白质相互作用分析(PPI);利用METASCAPE数据库进行核心靶点GO功能富集和KEGG通路富集分析。结果:川牛膝主要化学成分为槲皮素、杯苋甾酮、红苋甾酮、β-谷甾醇、齐墩果酸等;共筛选出关键共同靶点170个,包括AKT1、IL6、VEGFA、TP53、CASP3、TNF、JUN、EGF、MAPK1、EGFR、PTGS2、STAT3等;GO富集分析得到生物学进程主要涉及炎症反应、对伤害的反应、对脂多糖的反应、活性氧代谢过程、凋亡信号通路等;KEGG富集信号通路主要包含糖尿病并发症AGE-RAGE信号路径、肿瘤的途径、乙型肝炎、TNF信号通路、肿瘤中的蛋白质多糖等。结论:川牛膝治疗高血压存在多通路、多靶点的可能,揭示了川牛膝治疗高血压的潜在分子机制。Objective:Elucidate the mechanism of action of Cyathula officinalis Kuan for treating hypertension based on network pharmacology.Methods:The TCMSP database was used to obtain the principle active ingredients and target found in Cyathula officinalis Kuan and the OMIM,Drugbank,TTD,CTD and DISGENT databases were used to obtain the target genes involved in hypertension.We also used STRING platform to conduct“protein-protein interactional”(PPI)analyses on the intersection targets and then used the METASCPE database to perform GO functional enrichment analysis and KEGG pathway enrichment analysis on the intersection targets.Results:The main chemical components of Cyathula officinalis Kuan are Quercetin,Cyasterone,Rubrosterone,β-Sitosterol,Oleanolic acid,et al;A total of 170 intersection targets were obtained including AKT1,IL6,VEGFA,TP53,CASP3,TNF,JUN,EGF,MAPK1,EGFR,PTGS2,STAT3,et al;GO enrichment analysis found that the biological process were enriched primarily in inflammatory response,response to wounding,response to lipopolysaccharide,reactive oxygen metabolism process and apoptosis signal pathway,et al and KEGG enrichment signal pathway were enriched primarily in AGE-RAGE signal pathway of Complications of diabetes,tumor pathway,hepatitis Bm,TNF signal pathway,proteoglycans in tumors,et al.Conclusion:There is a possibility of multiple pathways and targets in the treatment of hypertension by Cyathula officinalis Kuan.This paper reveals the potential molecular mechanisms involved in the treatment of hypertension by Cyathula officinalis Kuan.
