摘 要:目的:探究升清降浊颗粒对脂多糖(LPS)致幼龄急性肺炎大鼠模型的保护作用及NLRP3蛋白的影响。方法:将50只雄性大鼠随机分空白组、模型组、升清降浊颗粒高、中、低剂量组,利用LPS尾静脉注射建立大鼠急性肺炎模型,造模后计算大鼠右肺组织的湿/干质量(W/D)比值,ELISA法检测血清和肺组织中IL-1β、TNF-α、IL-6炎症因子水平,Western Blot检测肺组织中NLRP3蛋白表达水平。结果:与空白组相比,LPS组大鼠的右肺组织W/D比值显著升高(P<0.01),而与给药组相比,模型组W/D显著性降低(P<0.01)。与空白组比较,模型组血清和肺组织中IL-1β、IL-6、TNF-α水平升高(P<0.01);与模型组相比,给药组血清和肺组织中IL-1β、IL-6、TNF-α水平均下降,其中高剂量升清降浊颗粒组最为明显。与空白组相比,模型组大鼠肺组织中NLRP3蛋白表达水平升高(P<0.01)。与模型组相比,给药组NLRP3蛋白表达降低,其中高剂量组TRPV1蛋白降低效果最明显(P<0.01)。结论:升清降浊颗粒对LPS致急性肺炎大鼠具有保护作用,作用机制可能与降低机体炎性因子产生和下调NLRP3信号蛋白有关。Objective:To investigate the protective effect of Shengqing Jiangturu Granules on lipopolysaccharide(LPS)-induced acute pneumonia in young rats and the effect of NLRP3 protein.Methods:Fifty male rats were randomly divided into blank group,model group,high,medium and low dose groups of Shengqing Jiangzhuo Granules.The rat model of acute pneumonia was established by tail vein injection of LPS.The wet/dry weight(W/D)ratio of right lung tissue was calculated after modeling.The levels of IL-1β,TNF-α and IL-6 inflammatory factors in serum and lung tissue were detected by ELISA.The expression of NLRP3 protein in lung tissue was detected by Western Blot.Results:Compared with the blank group,the W/D ratio of the right lung tissue of the LPS group was significantly increased(P<0.01),while compared with the administration group,the W/D ratio of the model group was significantly decreased(P<0.01).Compared with the blank group,the levels of IL-1β,IL-6 and TNF-α in serum and lung tissue of the model group were increased(P<0.01).Compared with the model group,the levels of IL-1β,IL-6 and TNF-α in serum and lung tissue of the administration group decreased,and the high-dose Shengqing Jiangzhuo Zhidong granule group was the most obvious.Compared with the blank group,the expression level of NLRP3 protein in the lung tissue of the model group was increased(P<0.01).Compared with the model group,the expression of NLRP3 protein in the administration group was decreased,and the decrease of TRPV1 protein in the high-dose group was the most obvious(P<0.01).Conclusion:Shengqing Jiangzhuo Granule has a protective effect on LPS-induced acute pneumonia in rats,the mechanism of which may be related to reducing the production of inflammatory factors and down-regulating NLRP3 signaling protein.
