摘 要:为了鉴定禾谷镰孢中KP关键酶基因及其功能,本研究采用生物信息学方法对禾谷镰孢的KP关键酶基因进行了系统分析。研究发现,禾谷镰孢含有KP关键酶IDO(吲哚-2,3-双加氧酶)、AFMID(芳基甲酰胺酶)、KAT(犬尿氨酸氨基转移酶)、KMO(犬尿氨酸单加氧酶)、KYN(犬尿氨酸酶)和HAO(羟基酸氧化酶)。通过分析禾谷镰孢的表达谱数据,发现KP关键酶基因FgIDO、FgAFMID、FgKAT、FgKMO、FgKYN和FgHAO在分生孢子发育、菌丝生长和侵染玉米茎秆的不同阶段表现出较高的表达水平,表明这些基因可能参与分生孢子发育、菌丝生长和侵染过程。整合研究结果和现有文献信息,绘制了禾谷镰孢KP及其关键酶催化反应的模式图,为阐明禾谷镰孢KP调控病菌生长发育和致病力的功能与机制奠定了基础。In order to identify the KP key gene and its function,the KP key enzyme gene of Fusarium gramineum was systematically analyzed by bioinformatics method.The key KP enzymes were identified in F.gramineum including IDO(indole-2,3-dioxygenase),AFMID(Arylformamidase),KAT(Kynurenine aminotransferase),KMO(Kynurenine monooxygenase),KYN(Kynureninase)and HAO(hydroxyl acid oxidase).Transcriptom data of F.graminearum showed that the key KP enzyme genes FgIDO,FgAFMID,FgKAT,FgKMO,FgKYN and FgHAO of F.graminearum were highly expressed during conidium stage,mycelium growth stage and different stages of maize stalk infection.The results indicated that the KP genes FgIDO,FgAFMID,FgKAT,FgKMO,FgKYN and FgHAO were involved in conidial production,mycelial growth and infection.Then the pattern map of KP and its key enzyme catalytic reaction of F.graminearum was drawn after summarization of the results and the existing literature information,which laid a foundation for elucidating the function and mechanism of KP key enzyme regulating the growth and pathogenicity of F.graminearum.
