摘 要:目的:探讨中华跌打丸对大鼠急性软组织损伤的保护作用及机制。方法:采用重物高空垂直下落造成打击伤建立大鼠急性软组织损伤模型。180只大鼠随机分为正常对照组,模型组,阳性药组,中华跌打丸高、中、低(2、1、0.5 g/kg)剂量组,每组30只。灌胃给予相应药物,给药第1、3、5天各组分别随机处死10只大鼠取材,测定肿胀度、压痛阈值、局部组织中的肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)、前列腺素E2(PGE2)水平和神经生长因子(NGF)含量,HE染色观察肌肉组织病理并进行评分。结果:与模型组比较,中华跌打丸各剂量组均可显著增加压痛阈值(给药第1、3、5天)(P<0.05,P<0.01),降低损伤局部肿胀度及TNF-α、IL-1β、PGE2水平,增加NGF含量(给药第3、5天)(P<0.01,P<0.05),降低肌肉病变总评分(给药第5天)(P<0.05);中华跌打丸中、高剂量组可显著降低肌肉病变总评分(给药第3天)(P<0.05);中华跌打丸高剂量组还可以显著抑制PEG2水平,降低肌肉病变总评分,提高NGF含量(给药第1天)(P<0.05,P<0.01)。结论:中华跌打丸对急性软组织损伤大鼠具有明显保护作用,其作用机制与降低损伤局部的TNF-α、IL-1β、PGE2水平和升高NGF含量有关。Objective:To investigate the protective effect and mechanism of Zhonghua Dieda Pills on rats with acute soft tissue injury.Methods:A model of acute soft tissue injury in rats was established by using the vertical fall of heavy objects from high altitudes to cause soft tissue injuries.A total of 180 rats were randomly divided into normal control group,model group.positive drug group,and high,medium and low(2,1,0.5 g/kg)dose groups of Zhonghua Dieda Pills,with 30 rats in each group.On the Ist,3rd and 5th days of giving the corresponding drugs by gavage,l0 rats were randomly sacrificed in each group,and the swelling degree,tenderness threshold,tumor necrosis factorα(TNF-a),interleukin 1β(IL-1β),prostaglandin E,(PGE,)and nerve growth factor(NGF)in local tissues were measured,and the pathology of muscle tissue was observed and scored by HE staining.Results:Compared with the model group,each dose group of Zhonghua Dieda Pills could significantly increase the tenderness threshold(on the 1st,3rd and 5th days of administration)(P<0.05.,P<0.01),reduce the local swelling and TNF-α,IL-1βand PGE2,increase the NGF(on the 3rd and 5th days of administration)(P<0.01,P<0.05),and reduce the total score of muscle lesions(on the 5th day of administration)(P<0.05).The medium-and high-dose groups of Zhonghua Dieda Pills could significantly reduce the total score of muscle lesions(on the third day of administration)(P<0.05).The high-dose group of Zhonghua Dieda Pills could also significantly inhibit PEG,level,reduce the total score of muscle lesions,and increase the content of NGF(on the first day of administration)(P<0.05,P<0.01).Conclusion:Zhonghua Dieda Pills have obvious protective effect on the acute soft tissue injury of rat model,and their mechanism is related to reducing the levels of TNF-α,IL-1β,PGE,and increasing the content of NGF in theinjured area.
