摘 要:目的探讨平喘宁基于磷脂酰肌醇3-激酶(PI3K)调控相关信号分子表达,对于卵清蛋白(OVA)致敏诱导的寒哮大鼠模型肺组织气道炎症的作用机制。方法将105只健康雄性SD大鼠随机分为7组,分别为正常组、模型组、平喘宁高剂量组、平喘宁中剂量组、平喘宁低剂量组、桂龙咳喘宁组、地塞米松组,每组15只。在寒冷环境下,以OVA对除正常组之外的其他组大鼠进行致敏,以建立模型。造模成功后,每日予以正常组和模型组等体积0.9%氯化钠溶液,平喘宁高、中、低剂量组分别予以平喘宁汤剂14.578、7.289、3.645 g/(kg·d)灌胃给药,桂龙咳喘宁组、地塞米松组分别予以桂龙咳喘宁片溶于0.9%氯化钠溶液的药液0.405 g/(kg·d)、地塞米松片溶于0.9%氯化钠溶液的药液0.405 mg/(kg·d)灌胃给药。采用苏木素-伊红(HE)染色法观察各组大鼠肺组织病理变化;酶联免疫吸附剂测定(ELISA)法检测各组大鼠支气管肺泡灌洗液(BALF)中白细胞介素(IL)-13、IL-25水平;反转录聚合酶链反应(RT-PCR)检测NADPH氧化酶4(NOX4)mRNA、维甲酸诱导基因Ⅰ(RIG-I)mRNA、胶原蛋白VⅠ(Col6)mRNA的表达水平。在整个实验过程中,对大鼠一般行为学状况进行观察和记录。结果HE染色法可见相较于正常组,模型组大鼠肺组织的平滑肌增生,有明显炎性细胞浸润,管壁增厚;相较于模型组,各治疗组大鼠肺组织病理情况均有改善。ELISA法可见相较于正常组,模型组大鼠BALF中IL-13、IL-25表达上升(P<0.01);相较于模型组,平喘宁高、中、低剂量组的IL-13、IL-25表达差异均有统计学意义(P<0.01)。RT-PCR法可见相较于正常组,模型组大鼠肺组织中NOX4、RIG-I、Col6 mRNA表达差异均有统计学意义(P<0.01);相较于模型组,各治疗组差异均有统计学意义(P<0.01)。大鼠一般行为学状况观察可见相较于正常组,模型组大鼠形体消瘦,皮毛枯槁,扎堆聚集,呼吸急促,打喷嚏,可闻及哮鸣声;�Objective To explore the mechanism of Pingchuanning(平喘宁)role in regulating the expression of related signaling molecules based on PI3K in lung tissue airway inflammation induced by ovalbumin(OVA)sensitization in a cold asthma rat model.Methods 105 healthy male SD rats were randomly divided into 7 groups,named as normal group,model group,Pingchuanning high,medium and low dose group,Guilong Kechuanning(桂龙咳喘宁)group,and dexamethasone group,with 15 rats in each group.In a cold environment,OVA was used to sensitize rats in groups other than the normal group to establish a model.After successful modeling,the normal group and model group were given equal volumes of physiological saline daily.Pingchuanning high,medium and low dose groups were given 14.578,7.289,and 3.645 g/(kg·d)respectively.Guilong Kechuanning group and dexamethasone group were given 0.405 g/(kg·d)and 0.405 mg/(kg·d)by respectively.Hematoxylin eosin(HE)staining was used to observe the pathological changes in lung tissue of rats in each group.Enzyme linked immunosorbent assay(ELISA)was used to detect interleukin-13(IL-13)and interleukin-25(IL-25)levels in BALF of rats in each group.Reverse transcription polymerase chain reaction(RT-PCR)was used to detect the expression of NOX4 mRNA,RIG-I mRNA and Col6 mRNA.Throughout the entire experimental process,the general behavioral status of rats was observed and recorded.Results HE staining method showed,compared with the normal group,smooth muscle hyperplasia in the lung tissue of the model group rats,with obvious inflammatory cell infiltration and thickening of the tube wall;compared with the model group,the pathological conditions of the lung tissue of rats in each treatment group improved.ELISA method showed,compared with the normal group,the expression of IL-13 and IL-25 in the BALF of the model group rats increased(P<0.01);Compared with the model group,there were statistical differences in the expression of IL-13 and IL-25 among the high,medium,and low dose groups of Pingchuanning(P<0.01).R
