摘  要:目的 探讨微型染色体维持蛋白6(MCM6)在皮肤黑色素瘤(SKCM)中的表达及其与预后的关系。方法 通过分析TCGA和GTEx数据库,比较MCM6在SKCM和正常皮肤组织中的表达差异;采用受试者工作特征曲线(ROC)、Cox回归分析和Kaplan-Meier (K-M)分析评估MCM6在SKCM中的临床意义及与预后之间的关系;通过LinkedOmics数据库、蛋白-蛋白相互作用(PPI)网络、GO/KEGG富集及免疫细胞浸润分析对与MCM6共表达的基因进行功能富集分析。结果 MCM6在SKCM中的表达水平明显高于正常皮肤组织(P<0.05),且MCM6的高表达与SKCM的不良预后密切相关。GO功能注释分析显示,与MCM6共表达基因主要参与细胞器分裂、DNA复制、细胞周期检查点等生物学过程。KEGG分析显示,这些基因在细胞周期、细胞衰老、p53信号途径及对药物的耐药性形成等信号传导过程中发挥着重要作用。免疫细胞浸润分析显示,MCM6表达水平与Th2细胞、辅助性T细胞的浸润水平呈正相关;与浆细胞样树突细胞、Th17细胞、自然杀伤细胞及细胞毒性T细胞的浸润水平呈负相关。结论 MCM6在SKCM的发生发展过程中发挥着重要作用,并且与SKCM的预后密切相关,可成为SKCM诊断和预后评估的潜在标志物。Objective To explore the expression of microchromosome maintenance complex 6(MCM6)in skin cutaneous melanoma(SKCM)and its relationship with the development and prognosis of SKCM.Methods The MCM6 mRNA expression profile data from The Cancer Genome Atlas(TCGA)and the Genotype-Tissue Expression(GTEx)databases was analyzed to compare the expression differences of MCM6 between SKCM and normal skin tissues.The correlation between MCM6 expression level and clinicopathological features and prognosis of SKCM patients was assessed by using Receiver Operating Characteristic(ROC)curves,Cox regression analysis,and Kaplan-Meier(K-M)survival analysis.Furthermore,functional enrichment analysis of genes co-expressed with MCM6 was conducted using the Linked Omics database,Protein-Protein Interaction(PPI)networks,Gene Ontology(GO)/Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis,and immune cell infiltration analysis.Results The expression level of MCM6 in SKCM was significantly higher than that in normal skin tissues(P<0.05),and its high expression was closely associated with poor prognosis of SKCM.The GO functional annotation analysis revealed that the co-expressed genes with MCM6 were primarily involved in biological processes such as organelle fission,DNA replication,and cell cycle checkpoints.And the KEGG analysis indicated that the co-expressed genes with MCM6 played crucial roles in signaling pathways related to cell cycle,cellular senescence,p53 signaling pathway,and drug resistance.The immune cell infiltration analysis demonstrated that the expression level of MCM6 was positively correlated with the infiltration levels of Th2 cells and helper T cells,but negatively correlated with The infiltration levels of plasma-like dendritic cells,Th17 cells,natural killer cells,and cytotoxic T cells.Conclusion MCM6 appears to play a pivotal role in the occurrence and development of SKCM,and is closely related to the prognosis of SKCM patients,making it a potential biomarker for SKCM treatment and prognosis evaluation.
