摘 要:为探究芍药2n雌配子人工诱导体系以获得四倍体后代,以芍药二倍体品种‘朱砂判’(Paeonia lactiflora‘Zhushapan’)为研究材料,通过石蜡切片法探索芍药大孢子及胚囊发生过程,建立芍药花蕾形态与大孢子及胚囊发生过程的关系。在此基础上,通过控制变量法探究秋水仙素质量浓度和注射次数对芍药2n雌配子诱导效率的影响。结果表明:芍药雌配子发育类型为蓼型,倒生胚珠,双珠被。芍药花蕾直径在26 mm≤d<29 mm范围,胚珠发育至二核胚囊主导期,质量浓度为3 g/L的秋水仙素注射花蕾3次,获得2个四倍体后代,2n雌配子诱导率最高,为3.33%。To explore the artificial induction system of 2n female gametes in Paeonia lactiflora to obtain tetraploid offspring, the diploid P. lactiflora variety ‘Zhushapan’ was used as the research material. The process of megasporogenesis and embryo sac development in P. lactiflora was investigated using paraffin sectioning, and the relationship between the flower bud morphology of P. lactiflora and the process of megasporogenesis and embryo sac development was established. Based on this, the effect of varying concentrations of colchicine and injection frequencies on the induction efficiency of 2n female gametes in P. lactiflora was explored using a controlled variable method. The results showed that the female gamete development type of P. lactiflora is polygonum-type, with inverted ovules and double integuments. When the flower bud diameter of P. lactiflora is in the range of 26 mm≤ d <29 mm and colchicine is injected into the buds three times with a concentration of 3 g/L during the dominance of the two-nuclear embryo sac stage, two tetraploid offspring were obtained. The induction rate of 2n female gametes was highest at 3.33%.
