摘  要:目的:建立HPLC双波长法同时测定甘草中甘草苷、甘草素、异甘草苷、异甘草素和甘草酸含量,为甘草质量的综合评价提供参考。方法:采用Zorbax SB-C18色谱柱(250 mm×4.6 mm,5μm),以0.1%磷酸-乙腈为流动相,梯度洗脱,检测波长为248 nm和362 nm,柱温为30℃,流速为1.0 mL/min。使用统计学软件SPSS 21.0对15批甘草的测定结果进行主成分分析。结果:5种活性成分分离效果良好,精密度、重复性、稳定性、线性关系、加样回收率和耐用性均良好。主成分分析可知,新疆甘草总体质量较好并且差异较小。结论:本法可同时测定5种成分的含量且可靠简便,结合主成分分析可为甘草质量的综合评价提供参考。Objective:To establish a dual-wavelength HPLC method for simultaneous determination of liquiritin,liquiritigenin,isoliquiritin,isoliquiritigenin and glycyrrhizic acid in glycyrrhiza,providing reference for comprehensive evaluation of the quality of glycyrrhiza.Methods:The HPLC separation was performed on a Zorbax SB-C18 column(250 mm×4.6 mm,5μm)with gradient elution of 0.1%phosphoric acid in water-acetonitrile,and peaks were detected at 248 nm and 362 nm,the column temperature was maintained at 30℃the flow rate was 1.0 mL/min,The determination results of 15 batches glycyrrhiza uralensis were analyzed by SPSS 21.0 with principal component analysis.Results:The five active ingredients were well separated with good precision,reproducibility,stability,linearity,spiked recoveries and durability.The principal component analysis showed that the overall quality of Xinjiang Glycyrrhiza glabra was good and the differences were small.Conclusion:The method can be used to determine the contents of the five components simultaneously,and it is reliable and simple,and combined with the principal component analysis,it can provide a reference for the comprehensive evaluation of the quality of Glycyrrhiza glabra.
