摘 要:硫化氢(H_(2)S)作为活性硫物质之一,其在生物体中参与多种生理过程如维持细胞氧化还原平衡、缺血再灌注损伤、抗炎和细胞凋亡等。因此,为了更好地了解硫化氢在生物系统中的作用和对疾病进行准确诊断,本文通过铃木偶联反应和取代反应合成了可用于高效检测的氧杂蒽结构的化合物1,并且详细地研究了其对硫化氢的荧光响应。化合物1本身发射出明亮的橙色荧光,与硫化氢响应后,荧光减弱甚至消失,并且对其他的阴离子和氨基酸的响应极其微弱。其验证机理已经通过高分辨质谱进行了验证。探针1识别H_(2)S具有良好的选择性和抗干扰能力,具有较低的检测限。最重要的是,探针已成功应用活细胞中外源性H_(2)S的荧光成像。Hydrogen sulfide(H_(2)S),as one of the living substances,participates in a variety of physiological processes in organisms,such as maintaining cell REDOX balance,ischemia-reperfusion injury,anti-inflammation and apoptosis.Therefore,in order to better understand the role of H_(2)S in biological systems and accurate diagnosis of diseases,compound 1 with the structure of oxacanthrene was synthesized by Suzuki coupling reaction and substitution reaction,and its fluorescence response to H_(2)S was studied in detail.Compound 1 itself emits bright orange fluorescence,which weakens or even disappears in response to hydrogen sulfide,and its response to other anions and amino acids is extremely weak.The validation mechanism has been verified by high-resolution mass spectrometry.Probe 1 recognized H_(2)S with good selectivity and anti-interference ability and low detection limit.Most importantly,the probe has been successfully applied for fluorescence imaging of exogenous H_(2)S in living cells.
