摘 要:为了解决聚丙烯酰胺造成的环境污染问题,通过富集、驯化从大庆油田含聚丙烯酰胺的污水中分离得到13株聚丙烯酰胺降解细菌,采用淀粉碘化镉分光光度法测定其降解率,从中筛选出3株降解率较高的细菌A1、E1和E4,经形态学和分子生物学鉴定为枯草芽孢杆菌(Bacillus subtilis)、黏质沙雷氏菌(Serratia marcescens)和产气克雷伯氏菌(Klebsiella aerogenes)。分别研究了聚丙烯酰胺的初始浓度、接菌量、不同碳氮源和金属离子等条件对复合菌A1E1E4降解聚丙烯酰胺的影响,并采用响应面法优化其降解条件。结果表明:复合菌A1E1E4以接菌量2%接种到聚丙烯酰胺的质量浓度为300.00 mg/L、NH_(4)Cl的质量浓度为569.48 mg/L、葡萄糖的质量浓度为709.16 mg/L、Ca^(2+)的浓度为3.57 mmol/L的基础培养液中,在温度为30℃条件下培养5 d,降解率达到61.45%。将复合菌A1E1E4加入到含聚丙烯酰胺油田污水中,70 d时降解率可达93.72%,比自然降解提高了70.84%。In order to solve the environmental pollution caused by PAM,13 polyacrylamide degrading bacteria were isolated from the waste water containing polymer in Daqing Oil Field by enrichment and domestication.The degradation rate was determined by starch cadmium iodide method,and three bacteria A1,E1 and E4 with high degradation rate were screened.They were identified as Bacillus subtilis,Serratia marcescens and Klebsiella aerogenes by morphology and molecular biology.The effects of initial concentration,inoculation amount,different carbon and nitrogen sources and metal ions on the degradation of polyacrylamide by A1E1E4 were studied,and the degradation conditions were optimized by response surface method.The results show that:The composite bacteria A1E1E4 were inoculated into the basic medium containing polyacrylamide with a mass concentration of 300.00 mg/L,NH_(4)Cl with a mass concentration of 569.48 mg/L,glucose with a mass concentration of 709.16 mg/L,and Ca^(2+)with a concentration of 3.57 mmol/L at the inoculation rate of 2%.Under the condition of 30℃for 5 days,the degradation rate reached 61.45%.When the complex bacteria were added into the oilfield containing PAM,the degradation rate reached 93.72%after 70 days,which was 70.84%higher than that of natural degradation.The results provide theoretical basis andtechnical support for bioremediation of PAM polluted environment.
