摘 要:目的:建立紫外-可见分光光度法测定灵芝菌丝中多糖含量的方法。方法:以无水葡萄糖为对照品,苯酚和硫酸为显色剂,在485nm波长处测定灵芝菌丝体中多糖含量。结果:灵芝菌丝中多糖含量0~0.11520 mg范围内线性关系良好(R2=0.9992),精密度RSD为1.42%。样品溶液在配制后12 h内稳定性较好,平均加标回收率为100.61%,RSD为0.97%。结论:该方法方便快捷,重现性好,结果可靠,适用于测定灵芝菌丝体中多糖含量。Objective:To investigate the applicability of UV-vis spectrophotometry for the determination of polysaccharide in Ganoderma lucidum mycelium.Methods:The contents of polysaccharide in ganoderma lucidum mycelia were determined at 485 nm with anhydrous glucose as reference substance,phenol and sulfuric acid as chromogenic agent.Results:The polysaccharide in ganoderma lucidum showed a good relationship with in the range of 0~0.11520 mg(R2=0.9992),precision RSD was 1.42%.The sample solution has good stability within 12 hour after preparation,the average recovery rate was 100.61%,RSD was 0.97%.Conclusion:The method was convenient and fast.It has good reproducibility and reliable results.It can be used to determine the content of polysaccharide in ganoderma lucidum mycelium.
