摘 要:目的探讨miR-194对兔耳增生性瘢痕的治疗作用。方法自2022年8月至2023年8月,哈尔滨医科大学附属第一医院整形美容中心对6只新西兰大白兔建立兔耳瘢痕模型,通过注射miR-194对兔耳瘢痕进行治疗。通过大体观察、HE染色、MASSON染色检测miR-194对兔耳瘢痕形态的影响。蛋白免疫印迹实验检测miR-194对于TGF-β1,VEGF和COL-1的表达的作用。ELISA检测miR-194对于TGF-β1、IL-1β和TNF-α的表达的作用。结果大体观察显示,治疗组的瘢痕更小更软且更平整,颜色也更接近正常皮肤颜色。HE染色切片显示,miR-194治疗可以降低兔耳瘢痕的形成,成纤维细胞排列更为平整、下调炎症因子的释放以及血管的新生。蛋白免疫印迹实验结果显示,miR-194能够明显抑制TGF-β1、VEGF和COL-1的表达。ELISA结果表明,miR-194能够降低TGF-β1、IL-1β和TNF-α的表达(P=0.02,P=0.01,P=0.02)。结论miR-194能够通过降低组织中TGF-β1,VEGF和COL-1的表达,下调炎症因子的活性发挥抑制增生性瘢痕形成的作用。Objective To explore the therapeutic effect of miR-194 on hypertrophic scars in rabbit ears.Methods From August 2022 to August 2023,the Plastic Surgery and Cosmetic Center of the First Affiliated Hospital of Harbin Medical University established a rabbit ear scar model on 6 New Zealand white rabbits,and treated the rabbit ear scars by injecting mi R-194.Detect the effect of mi R-194 on the morphology of rabbit ear scars through gross observation,HE staining,and MASSON staining.Protein immunoblotting assay was used to detect the effects of mi R-194 on the expression of TGF-β1,VEGF,and COL-1.ELISA detection of the effect of miR-194 on the expression of TGF-β1,IL-1β and TNF-α.Results General observation showed that the scars in the treatment group are smaller,softer,and smoother,with a color closer to normal skin color.HE staining sections showed that mi R-194 treatment can reduce the formation of rabbit ear scars,flatten the arrangement of fibroblasts,downregulate the release of inflammatory factors,and promote angiogenesis.The results of protein immunoblotting experiments showed that mi R-194 can significantly inhibit the expression of TGF-β1、VEGF and COL-1.The ELISA results showed that miR-194 could reduce the expression of TGF-β1、IL-1β and TNF-α(P=0.02,P=0.01,P=0.02).Conclusion Mi R-194 can inhibit the formation of hypertrophic scars by reducing the expression of TGF-β1,VEGF,and COL-1 in tissues and downregulating the activity of inflammatory factors.
