摘 要:为了实现海产品体内氧化三甲胺(trimethylamineoxide,TMAO)的快速定量检测,提出一种采用微流控光化学还原芯片进行前处理的电化学分析方法.首先构建了PDMS/SiO_(2)微流控芯片,并对TMAO的前处理反应条件进行了优化:采用该微流控芯片,使用Fe(Ⅱ)-HEDP为还原剂,在pH值为6.5,温度为40℃,紫外光照时间为2min的条件下,TMAO被还原为三甲胺(trimethylamine,TMA)的还原率达99.44%;然后,利用自制选择性电极对TMA溶液进行检测,在0.01~10mmol·L^(-1)范围内,TMA浓度对数与其溶液电位值线性相关,相关系数r^(2)=0.9977,检测限为2.28μmol·L^(-1).采用该方法对实际海鲜产品进行TMAO检测的相对偏差不超过4.71%,加标回收率为91.78%~112.16%.该方法测定时间短、结果准确,在海产品分析检测中具有良好的应用前景。Aiming at the rapid quantitative detection of trimethylamine oxide(TMAO)in seafood,an electrochemical analysis method using microfluidic photochemical reduction chip for pretreatment is proposed.Firstly,the PDMS/SiO,hybrid microfluidic chip is fabricated,and the pre-treatment reaction reduction conditions of TMAO are optimized.Using this microfluidic chip and Fe(Ⅱ)-HEDP as reducing agent,under the conditions of pH 6.5,temperature 40'C and UV-irradiation time 2 min,the reduction rate of TMAO to trimethylamine(TMA)can reach 99.44%.Then,the self-made selective electrode is used to detect TMA solution.In the range of 0.01~10 mmol·L^(-1),the logarithm of TMA concentration is linearly correlated with the solution potential value,the cor-relation coefficient r^(2) is 0.9977,and the detection limit is 2.28μmol·L^(-1).The relative deviation of this method for TMAO detection of actual seafood products is not more than 4.71%,and the recovery rate is 91.78%~112.16%.The method has the advantages of short determination time and accurate results,and shows a good application prospect in the analysis and detection of seafood.
