摘 要:目的:建立蒙药材蜀葵花的定性定量评价方法。方法:采用显微法、薄层色谱法建立蜀葵花的定性鉴别方法,2020年版《中国药典》方法测定水分、总灰分、酸不溶性灰分、浸出物、含量测定,采用高效液相色谱法建立蜀葵花中山柰素的定量分析方法。结果:建立的薄层色谱法斑点清晰,重复性好,可用于对7批蜀葵花药材的定性鉴别。测得7批样品水分、总灰分、酸不溶性灰分、浸出物含量。蜀葵花中山柰素含量测定的标准曲线为Y=0.0471X-0.0288,线性范围为10.81ng~135.13 ng,相关系数为0.9999,精密度RSD=0.87%,加样回收率为102.03%,RSD=1.82%。7批蜀葵花中山柰素含量为0.12%~0.49%。结论:建立的显微法、薄层色谱法可用于对蜀葵花药材的定性鉴别,高效液相色谱法简便、可行,可用于蜀葵花中山柰素的含量测定。Objective:To establish a qualitative and quantitative evaluation method for the medicinal materials of hollyhock.Methods:The qualitative identification method of hollyhock flower was established by microscopy and thin layer chromatography,The 2020 edition of the"Chinese Pharmacopoeia"method for the determination of moisture,total ash,acid-insoluble ash,and extract content,and the quantitative analysis method of kaempferol in hollyhock flower was established by HPLC.Results:The established TLC had clear spots and good repeatability,which could characteristically realize the qualitative identification of 7 batches of hollyhock.Measured the moisture,total ash,acid-insoluble ash,and extract content of 7 batches of samples.The standard curve for the determination of kaempferol in hollyhock is Y=0.0471X-0.0288,the linear range is 10.81ng~135.13 ng,the correlation coefficient is 0.9999,the precision RSD=0.87%,the average sample recovery rate is 102.03%,RSD=1.82%.The content of kaempferol in 7 batches of hollyhock flowers was 0.12%~0.49%.Conclusion:The established microscopic method and thin-layer chromatography can realize the qualitative identification of hollyhock medicinal materials.High performance liquid chromatography is simple,feasible,and can be used for the content determination of kaempferol in hollyhock.
