摘 要:目的:比较野百合碱(MCT)、低压低氧(HPH)、Su5416联合低压低氧(SuHx)3种方法构建肺动脉高压大鼠模型的特点。方法:雄性SD大鼠随机分为对照组和3个实验组:MCT组、HPH组、SuHx组,MCT组腹腔注射MCT 50 mg/kg后常规饲养4周,HPH组置于海拔高度为5000 m的低压氧舱8周,SuHx组皮下注射SU541620 mg/kg后置于海拔高度为5000 m的低压氧舱3周。造模结束后检测各组大鼠肺动脉压和右心室压、左心室功能、血细胞计数,计算大鼠心脏系数和右心肥厚指数,观察肺组织病理改变。结果:与对照组比较,实验组大鼠肺动脉压、右心室压、右心肥厚指数均升高,SuHx组右心室压升高较其他2个实验组更为显著,差异均具有统计学意义(P<0.05);HPH组、SuHx组左心室舒张功能障碍,红细胞和血红蛋白增多,血小板减少;而MCT组血小板增多,红细胞和血小板变化不大;与对照组比较,实验组肺动脉壁厚度明显增加,差异有统计学意义(P<0.05)。结论:3种方法均可构建肺动脉高压大鼠模型,其中SuHx造模效果最好。Objective:To compare the characteristics of three methods:monocrotaline(MCT),hypobaric hypoxia(HPH),and SU5416 combined with hypobaric hypoxia(SuHx)in constructing pulmonary arterial hypertension(PH)rat models.Methods:Male SD rats were randomly divided into a control group and three experimental groups:MCT group,HPH group,and SuHx group.The MCT group received intraperitoneal injection of MCT 50 mg/kg and was routinely raised for 4 weeks.The HPH group was placed in a low-pressure oxygen chamber at an altitude of 5000 meters for 8 weeks,and the SuHx group received subcutaneous injection of SU541620 mg/kg and placed in a low-pressure oxygen chamber at an altitude of 5000 meters for 3 weeks.After the model being made,the pulmonary artery pressure,right ventricular pressure,left ventricular function and blood cell count of rats in each group were measured,heart coefficient and the right heart hypertrophy index of rats were calculated,and the pathological changes of lung tissue were observed.Results:Compared with the control group,the pulmonary artery pressure,right ventricular pressure and right ventricular hypertrophy index of the experimental group rats increased,and the right ventricular pressure of the SuHx group increased more significantly than the other two experimental groups,the difference was statistically significant(P<0.05);The left ventricular diastolic function decreased,red blood cells and hemoglobin increased,and platelets decreased in the HPH and SuHx groups;However,in the MCT group,there was an increase in platelets,with little change in red blood cells and platelets;Compared with the control group,the pulmonary artery wall thickness of the experimental group significantly increased,the difference is statistically significant(P<0.05).Conclusion:Three methods can be used to construct a rat model of pulmonary hypertension,and SuHx has the best modeling effect.
