摘 要:目的 探讨肺宁汤对急性放射性肺炎模型大鼠炎症因子Th17/Treg平衡及NF-κB通路的影响。方法 40只SD大鼠随机分为空白组、模型组、试验组和阳性组,每组10只,复制SD大鼠急性放射性肺炎模型,空白组和模型组给予生理盐水灌胃,试验组和阳性组分别给予肺宁汤和地塞米松灌胃。检测血清IFN-γ、TNF-α和IL-6水平,Th17/Treg细胞含量,肺组织NF-κB通路p65和IκBα水平。结果 模型组IFN-γ、TNF-α和IL-6水平升高;试验组和阳性组IFN-γ、TNF-α和IL-6水平降低(P<0.05)。模型组Th17细胞升高,Treg细胞降低;试验组和阳性组Th17细胞降低,Treg细胞升高,且试验组优于阳性组(P<0.05)。模型组p65蛋白表达水平升高,IκBα蛋白表达水平降低;试验组和阳性组肺组织p65蛋白表达水平降低,IκBα蛋白表达水平升高,且试验组优于阳性组(P<0.05)。结论 肺宁汤保护肺组织的机制与降低炎症因子,调整Th17/Treg平衡及抑制NF-κB通路有关。Objective To investigate the effects of Feining decoction on cytokines,Th17/Treg balance in peripheral blood and NF-κB pathway in rats with acute radiation pneumonia.Methods Forty SD rats were randomly divided into blank group,model group,experimental group and positive group,with 10 rats in each group.The acute radiation pneumonia model of SD rats was duplicated.The blank group and model group were given saline by gastric lavage,the experimental group was given Feining decoction by gastric lavage,and the positive group was given dexamethasone.Two weeks later,lung tissues and blood samples were collected.HE staining was used to observe the pathological morphology of lung tissues.The levels of serum interferon-gamma(IFN-γ),tumor necrosis factor-alpha(TNF-α)and interleukin-6(IL-6)were detected.The content of Th17/Treg cells in peripheral blood was detected.The expression of p65 and inhibition of NF-κB pathway and Protein(IκBα)level in lung tissues were detected.Results The levels of IFN-γ,TNF-αand IL-6 in model group were increased,while the levels of IFN-γ,TNF-αand IL-6 in test group and positive group were decreased(P<0.05).Th17 cells in peripheral blood of model group increased,Treg cells decreased,Th17 cells in experimental group and positive group decreased,Treg cells increased,and the experimental group was better than the positive group(P<0.05).The expression level of p65 protein in model group was increased,and the expression level of I-κB-αprotein was decreased.The expression level of p65 protein in experimental group and positive group was decreased,and the expression level of I-κB-αprotein was increased,and the experimental group was better than the positive group(P<0.05).Conclusion Fengning decoction can protect lung tissue,and its mechanism is related to reducing inflammatory cytokines,adjusting Th17/Treg cell balance and inhibiting the expression of NF-κB pathway protein.
