摘 要:目的 调查昭通市宿主动物中鼠疫噬菌体的携带情况,对部分噬菌体进行初步鉴定。方法 2019年5月在昭通市永善、水富、绥江3个县采用鼠笼法及5 m夹线法捕获宿主动物,经鉴定采集肠道样本,采用双层琼脂平板法分离鼠疫噬菌体,对分离结果进行统计描述和分析。结果 本次调查共捕获宿主动物449只,分离到15株鼠疫噬菌体,总分离率为3.34%。有8个乡(镇)分离到鼠疫噬菌体,其中青胜乡分离率最高为16%(4/25)。不同生境、性别和年龄鼠疫噬菌体的分离率差异无统计学意义;不同种属宿主动物鼠疫噬菌体的分离率差异具有统计学意义。所分离噬菌体噬菌斑表现为3种类型,电镜下观察为肌尾病毒科噬菌体。结论 云南省昭通市存在鼠疫噬菌体,所分噬菌体为肌尾病毒科噬菌体且具有多态性。Objective To investigate whether Yersinia pestis phage are carried in host animals in Zhaotong City,and identify isolated plague phage.Methods In May 2019,host animals were captured by squirrel cage method and 5-meter clamp method in Yongshan,Shuifu and Suijiang counties of Zhaotong.Intestinal samples were collected after identification,Yersinia pestis phages were isolated by double-layer agar plate method,and isolation results were statistically described and analyzed.Results A total of 449 host animals were collected,from which 15 strains of Yersinia pestis phage were separated,with a total isolation rate of 3.34%.The Yersinia pestis phages were isolated in 8 villages,and the highest rates was 16%(4/25)in Yongshan Town.There was no significant difference in the isolation rate of pestis phages among different habitats,sexes and ages.The isolation rate of pestis bacteriophages from different species of host animals was statistically significant.The plaque of the phage is divided into three types,two phages are identified as the Myoviridae family.Conclusion The Yersinia pestis phages exist in Zhaotong,these phages are polymorphic phages and of the Myoviridae family.
