摘 要:髓细胞组织增生蛋白2(myelocytomatosis protein 2,MYC2)作为MYC型bHLH转录因子家族成员,是茉莉酸响应途径的关键转录因子,在调控植物抵抗逆境胁迫中具有重要作用。本研究基于NCBI数据库中野草莓(Fragaria vesca)的基因序列,从草莓(Fragaria×ananassa)品种‘红颜’(‘Benihoppe’)中克隆鉴定了1个FaMYC2-like基因,其开放阅读框长度为1473 bp,编码490个氨基酸残基。保守结构域分析表明,FaMYC2-like蛋白具有bHLH-MYC家族保守结构域。系统发育分析显示,FaMYC2-like蛋白与月季花(Rosa chinensis)等蔷薇科(Rosaceae)植物中的同源基因编码的蛋白质具有较近的亲缘关系。通过启动子顺式作用元件预测,发现其启动子区含有大量的光信号、胁迫响应及激素信号的响应元件。亚细胞定位结果表明,FaMYC2-like蛋白定位于细胞核中。组织特异性RT-qPCR结果显示,FaMYC2-like基因在草莓的根中表达量最高,在茎、叶和花中也有较高表达,在匍匐茎中表达量最低;在果实发育早期的小绿期和大绿期中表达量最高,在果实发育的中后期几乎不表达。低温、干旱和盐胁迫均能在早期显著诱导草莓根和叶中FaMYC2-like的表达。Myelocytomatosis protein 2(MYC2),as a member of MYC-type bHLH transcription factor family,is a key transcription factor in the jasmonate response pathway and plays an important role in regulating plant resistance to stress.Based on the sequence from wild strawberry(Fragaria vesca)in NCBI database,a FaMYC2-like gene was cloned and identified from strawberry(Fragaria×ananassa)‘Benihoppe'.The length of its open reading frame was 1473 bp,encoding 490 amino acid residues.Based on the conserved domains analysis,FaMYC2-like protein had a conserved domain of bHLH-MYC family.Phylogenetic analysis showed that FaMYC2-like protein was closely related to the homologous genes in mostly Rosaceae plants such as Chinese rose(Rosa chinensis).Through the prediction of cis-acting elements of the promoter,it was found that the promoter region contained a large number of response elements of light signal,stress response and hormone signal.The results of subcellular localization showed that FaMYC2-like protein was mainly located in the nucleus.Result of tissue-specific RT-qPCR showed that the expression of FaMYC2-like gene was highest in the root of strawberry,next was in stem,leaf and flower,with the lowest in the stolon.In different stages of fruit development,FaMYC2-like showed the highest expression in the small green stage and large green stage of early fruit development,and almost no expression in the middle and late fruit development.Low temperature,drought and salt stress all significantly induce the expression of FaMYC2-like in strawberry roots and leaves at early stage.
