摘 要:目的:观察生血通便颗粒对血虚肠燥型慢传输型便秘模型大鼠肠道运动、Cajal间质细胞(ICC)超微结构及酪氨酸激酶受体(c-Kit)表达的影响。方法:选取45只SPF级雄性SD大鼠分为空白组、模型组、中药组、西药组,除空白组外其余3组采用盐酸洛哌丁胺经胃灌服结合尾部放血法构造血虚肠燥型大鼠模型,经鉴定模型复制成功后,各实验组分别给予相应药物干预14天;观察记录各组大鼠治疗前后一般情况及相关指标追评变化。结果:与模型组比较,中药组及西药组大鼠一般情况明显改善,体质量、粪便含水率及肠道推进率明显升高(P﹤0.05),结肠ICC整体结构恢复或轻度异常,自噬小体减少,结肠组织c-kit mRNA及蛋白表达增加(P﹤0.05)。结论:生血通便颗粒可有效改善血虚肠燥型慢传输型便秘大鼠肠道动力,其机制可能与上调结肠组织c-kit表达、抑制ICC自噬、改善ICC超微结构有关。Objective:To approach the effects of Shengxue Tongbian granules on intestinal motility,ultrastructure of interstitial cells of Cajal(ICC)and c-Kit expression in slow transit constipation(STC)model rats with syndrome of intestine dryness due to deficiency of blood.Methods:45 SPF male SD rats were selected and divided into blank group,model group,TCM group and western medicine group.Except for blank group,other three groups were established into STC rat models with syndrome of intestine dryness due to deficiency of blood by gastric perfusion of loperamide hydrochloride and tail blood-letting.After the identified models were successfully copied,each experimental group was intervened with corresponding drug for 14 days.The changes of general conditions and the level of related indicators of rats in each group were observed and recorded before and after treatment.Results:Compared with model group,the general conditions of rats in TCM group and western medicine group was obviously improved;the body mass,fecal water content and intestinal propulsion rate were significantly increased(P<0.05);the overall structure of colon ICC was recovered,or slight abnormality;the autophagosomes decreased;the expressions of c-kit mRNA and protein in colon tissue increased(P<0.05).Conclusion:Shengxue Tongbian granules can effectively improve the intestinal motility of STC rats with syndrome of intestine dryness due to deficiency of blood.Its mechanism may be related to up-regulating the expression of c-kit in colon tissue,inhibiting ICC autophagy and improving ICC ultrastructure.
