摘 要:目的目前针对牙槽骨不规则缺损的修复尚无行之有效的治疗手段。制备一种包裹牙周膜干细胞和二甲双胍的磷酸钙(CPC)-微纤维(aMF)支架,评价其理化性能以及体外成骨效果,并分析其用于临床进行骨缺损修复的可能性。方法将磷酸四钙与无水碳酸氢钙混合制备磷酸钙骨水泥,将藻酸钠在7.5%的氧化条件下降解,将上述支架材料混合后,包裹牙周膜干细胞和二甲双胍,制备出CPC-aMF骨缺损修复支架。通过扫描电镜以及光学显微镜,对该支架进行初步理化性能和表面形貌的表征。最后通过碱性磷酸酶等成骨分化实验确定该支架的成骨效果。结果在该支架的表面形貌表征中发现,CPC提供了支撑结构,aMF作为其内包裹的细胞和生长因子保护。这种新型支架显示了良好的成骨性能,碱性磷酸酶半定量检测发现包裹牙周膜干细胞和二甲双胍的CPC-aMF支架的成骨效果是对照组的2.5倍,qT-PCR检测结果发现实验组的成骨效果是对照组的2.5倍。结论新型的骨缺损支架为治疗大面积骨缺损提供了一种潜在的治疗性骨组织工程策略。Objective There is no effective treatment method for now to repair irregular alveolar bone defects.We constructed a calcium phosphate cement(CPC)-alginate microfibers(aMF)scaffold encapsulating human periodontal ligament stem cells(hPDLSCs)and metformin for the first time.We evaluated its physicochemical properties and in vitro osteogenic effect,and analyzed its potential for clinical bone defect repair.Methods Calcium phosphate cement was prepared by mixing tetracalcium phosphate with anhydrous calcium bicarbonate.Sodium alginate was degraded under 7.5%oxidation conditions.After mixing the CPC and aMF scaffold materials,hPDLSCs and metformin were then encapsulated to construct the bone repairing scaffold.Preliminary characterization of the physicochemical properties and surface morphology of the scaffold were carried out using scanning electron microscopy and optical microscopy.Finally,the osteogenic effect of the scaffold was determined through osteogenic differentiation experiments such as alkaline phosphatase.Results It was found in the surface morphology characterization of the scaffold that CPC provided a supporting structure,and aMF provided protection for encapsulated cells and growth factors.This novel scaffold demonstrated good osteogenic effects.Semi-quantitative alkaline phosphatase testing found that the osteogenic effect of the CPC-aMF scaffold encapsulating hPDLSCs and metformin was 2.5 times higher than that of the control group.The qT-PCR test results also showed that the osteogenic effect of the experimental group was 2.5 times higher than that of the control group.Conclusion This novel CPC-aMF scaffold provided a potential therapeutic bone tissue engineering strategy for grafting bone fenestration and bone dehiscence patients.
