摘 要:为了为鸭疫里默氏菌与机体的相互作用及作用机制研究提供基础数据,以雏鸭为研究对象,开展鸭人工感染鸭疫里默氏菌试验,分别测定感染组和对照组1、2、3、5、9、14 d等不同时间段的体质量、小肠各肠段的长度以及血浆中内毒素和血清中乳酸脱氢酶含量;荧光定量PCR法检测肠黏膜屏障功能相关基因MUC2和MYLK的表达量,探讨鸭疫里默氏菌感染对鸭生长发育和肠黏膜屏障功能的影响。结果表明,与对照组相比,感染组5、9 d体质量显著降低;感染鸭疫里默氏菌5 d十二指肠和空肠的长度显著降低;感染鸭疫里默氏菌1 d血浆中内毒素和血清中乳酸脱氢酶的含量均显著增加;鸭人工感染鸭疫里默氏菌的MUC2和MYLK表达量在十二指肠和空肠主要表现为下调,而在回肠MUC2的表达量表现为先下降再上调,MYLK的表达量表现为先下调再恢复至对照水平。综上,鸭人工感染鸭疫里默氏菌会减缓鸭的体质量增长和小肠生长,引起鸭的肠黏膜屏障功能相关基因的表达发生变化。To provide basic data for the study on the interaction and mechanism of action between Riemerella anatipestifer and the body,in this study,taking ducklings as the research object,experiments on artificial infection of duck with Riemerella anatipestifer were conducted.Body weight,the length of each intestinal segment,and the content of endotoxin and lactate dehydrogenase in blood of infection control and control groups at different time periods of 1,2,3,5,9,and 14 days were measured,the expression of MUC2 and MYLK genes related to intestinal mucosal barrier function were detected by fluorescent quantitative PCR,and the effects of Riemerella anatipestifer infection on the growth and development and intestinal mucosal barrier function of ducks were investigated.The results showed that the body weight of 5 and 9 days of Riemerella anatipestifer infection was significantly lower than that of the control group.The length of duodenum and jejunum of 5 days of Riemerella anatipestifer infection was significantly lower than that of the control group.The contents of endotoxin and lactic acid in the blood of the Riemerella anatipestifer group were significantly increased on 1 day of Riemerella anatipestifer infection.The level of MUC2 and MYLK genes in duodenum and jejunum were decline,while the level of MUC2 in ileum decreased first and then increased,and that of MYLK decreased first and then returned to a level similar to control group.In conclusion,artificial infection of Riemerella anatipestifer could slow down body weight increase and small intestine growth,and cause changes in the expression of genes related to intestinal mucosal barrier function of ducks.
