摘 要:目的:研究生石膏煎煮工艺对清肺排毒汤微粒体系理化性质的影响。方法:以生石膏的不同煎煮方式(直接煎、先煎、包煎、另煎)制备清肺排毒汤水煎剂及配方颗粒调配液,以830×g、3300×g、13200×g的离心力通过梯度离心法分离各水煎剂和配方颗粒调配液中的固体微粒,对微粒体的粒径、电位、形貌、元素分布及蛋白质、多糖含量进行表征。结果:各水煎剂组和颗粒剂组微粒体的粒径均随着离心力的增大而递减,且水煎剂组微粒体粒径均大于颗粒剂组。各水煎剂组和颗粒剂组微粒体的电位始终为负值,差异无统计学意义。水煎剂的制备方式与生石膏煎煮方式显著影响了清肺排毒汤微粒体形貌,生石膏直接煎和另煎组水煎剂微粒体呈现大小不一的球形和椭球形团聚形态,其中分散着片状和纤维状物质;生石膏先煎和包煎组水煎剂微粒体呈现网状结构;颗粒剂微粒体表呈不规律的块状堆积状态。各微粒体的主要元素为C、O和Ca,多糖含量高于蛋白质,但水煎剂组微粒体中多糖的含量均低于颗粒剂组微粒体。结论:共煎共煮形成的清肺排毒汤微粒体的粒径、电位、形貌、元素分布及蛋白质、多糖含量与配方颗粒调配液均不同,并且生石膏的煎煮工艺也会影响水煎剂微粒体的理化性质及物质形态。Objective:To study the effect of gypsum decocting process on the physicochemical properties of the microparticle system of Qingfei Paidu Decoction.Methods:The water decoction and formula granule preparation solution of Qingfei Paidu Decoction were prepared by different decoction methods of gypsum(direct decoction,pre-decoction,wrapdecoction and separate decoction).The solid particles in each water decoction and formula granule preparation solution were separated by gradient centrifugation at centrifugal forces of 830×g,3300×g and 13200×g,respectively,and the particle size,potential,morphology,element composition,protein and polysaccharide contents were analyzed.Results:The particle size of the microparticles in each decoction groups and formula granule groups was decreased with the increase of centrifugal force,and the particle size of decoction groups was larger than that of granule groups.The potential of microparticles in each water decoction and formula granule groups was always negative,and there was no significant difference among the groups.The preparation method of water decoction and decocting method of gypsum significantly affected the morphology of the microparticles of Qingfei Paidu Decoction:the microparticles in gypsum direct decoction and separate decoction groups(water decoction)showed spherical and ellipsoidal shapes with different sizes,in which flaky and fibrous substances were observed;the microparticles in gypsum pre-decoction and wrap-decoction groups(water decoction)showed reticular structure;the microparticles in formula granule groups were accumulated as irregular bulk.The element composition of microparticles in each group is C,O and Ca,and the polysaccharide content was higher than that of protein.However,the content of polysaccharide in the water decoction groups was lower than that in the formula granule preparation solution groups.Conclusion:The particle size,potential,morphology,element composition,protein and polysaccharide contents of Qingfei Paidu Decoction prepared by"Co-deco
